The functional effects of different types of fibre on growth performance and gut health of piglets could be ascribed to their physicochemical properties, fermentation patterns and abilities to modify the microbiota. In order to establish a better strategy of fibre supplementation in diets, thirty piglets(approximately 21 ± 1 d age;7.50 ± 0.31 kg)were randomly allotted to 5 treatments, and fed Control diet(basal diet),IDF diet(1% insoluble fiber diet),SDF diet(1% soluble fiber diet),CRMDF diet(0.75% insoluble fibre + 0.25% soluble fibre in the first two weeks and 0.75% soluble fibre + 0.25% insoluble fibre in the last two weeks)and MDF diet(0.5% insoluble fibre + 0.5% soluble fibre diet). The exprement lasted 28 days.Feeding MDF diet decreased F/G(P < 0.05)in the first two weeks and diarrhea incidence(P < 0.05)in the last two weeks.CRMDF-and MDF-fed piglets had a lower(P < 0.05)F/G than that of Control-fed piglets in the whole period.Feeding CRMDF and MDF diets increased(P < 0.05)the serum TP and AL concentrations. All fibre groups showed a higher(P < 0.05)ATTD of DM,CP and OM than Control-fed piglets.Feeding SDF diet increased(P < 0.05)the weight index of colon when compared with Control group.SDF-,CRMDF-and MDF-fed piglets had higher gene expressions involved in intestinal development(GLP-2,IGF-1R and TGF-β2)in the ileum and colon than Control-fed piglets(P < 0.05).All fibre groups decreased the activity of serum DAO in variable degree.Feeding CRMDF and MDF diets up-regulated(P < 0.05)ileal gene expressions involved in intestinal barrier functions(OCLN,CLDN-1,MUC1,IL-10).All fibre groups up-regulated(P < 0.05)gene expressions of ZO-1,OCLN,CLDN-1,MUC2 and the concentration of s Ig A,while down-regulated(P < 0.05)gene expressions of IL-1β and TNF-α in colon in variable degree. The alpha diversity in SDF group were significantly higher than those in other groups(P < 0.05).At the phyla level, feeding IDF diet significantly increased the abundance of Bacteroidetes and Euryarchaeota(P < 0.05).The abundance of Proteobacteria and Actinobacteria were significantly higher in SDF group(P < 0.05).The abundance of Firmicutes was increased significantly in CRMDF group(P < 0.05).Piglets from MDF group had a higher abundance of Bacteroidetes(P < 0.05).At the genera level,IDF group significantly increased the abundance of Phascolarctobacterium,Coprococcus_1 and Prevotella_1(P < 0.05).Blautia,Syntrophococcus,Atopobium and Succinivibrio were significantly higher in SDF group(P < 0.05).In CRMDF group, the abundance of Blautia,Subdoligranulum and Roseburia were significantly increased(P < 0.05).Selenomonas,Phascolarctobacterium and Alloprevotella were significantly enriched in MDF group(P < 0.05).Feeding SDF and CRMDF diets decreased the p H value of digesta in hindgut(P < 0.05).The concentrations of SCFAs in IDF,CRMDF and MDF group in caecum were significantly increased(P < 0.05),and feeding SDF diet increased(P < 0.05)the concentrations of SCFAs in colon. In summary, feeding insoluble fiber might be more preferable to promote the growth performance and nutrients digestibility of piglets than soluble fibre in the first two weeks after weaning. In the whole period, diets with a combination of the two fibres had better effects on growth performance, serum biochemistry, nutrients digestibility, gene expressions involved in small intestinal development and barrier functions.SDF diet could increase alpha diversity of microbiota and concentrations of SCFAs,meanwhile, promote intestinal development and barrier functions in colon.
