Paris polyphylla Smith var.yunnanensis(Franch.)Hand.-Mazz.Symb.Sin.belongs to the Melanthiaceae family.The active ingredient of P.polyphylla var.yunnanensis is polyphyllin,which have the effects of analgesic,anti-inflammatory,hemostasis,inhibition of cancer cell and proliferation.Its rhizome can be used as medicine,and it is an important raw material for clinical medicine and pharmaceutical industry.According to statistics,the annual demand for rhizome of P.polyphylla var.yunnanensis and P.polyphylla var.chinesis is about 16,000 tons,and the annual output value of medicines and daily chemical products derived from these species is as high as 10 billion yuan.However,the slow seed breeding long cycle growth cannot meet the huge demanding market,which causing wild resources to be endangered.Tissue culture technology has become an effective method to solve the problem of P.polyphylla var.yunnanensis resources.In the past few years,we have successfully established a system of the tissue culture of P.polyphylla var.yunnanensis and selected some different strains.After transplanting the in vitro plantlets into the soil,it is necessary to adapt to the great changes in the environment outsides.In this paper,the acclimation of the in vitro plantlets of P.polyphylla var.yunnanensis was studied by detecting various morphological and physiological indexes after transplanting into soil.We distinguished differences in growth of ex vitro acclimated plantlets among different strains,and compared the active ingredients between acclimated plantlets and seedlings.For the first time,this paper describes the growth and development of the in vitro and ex vitro acclimated plantlets of the Paris polyphylla var.yunnanensis,and provides a practical assessment to rapproachable evaluation these strains.The results of the thesis are as follows:1.Some polyphyllins have been accumulated during the period of tissue culture in the bottles,and the survival rate after transplanting was 94%.2.After transplanting into the soil,the morphology of ex vitro acclimated plantlets were normal.As the acclimated time increased,their photosynthesis value,biomass accumulation and total active ingredients content increased.3.Different strains of in vitro plantlets produce more organic matter than the seedlings in the same time,just for their fresh weight storage.The content of polyphyllins in ex vitro acclimated plantlets was higher than that of the seedlings.4.The different strains of in vitro plantlets have the characteristics of rapid propagation,not only accumulate fresh weight,but also contain several kinds of polyphyllins prescribed by The Chinese Pharmacopoeia.Among these results,the rhizome of yu13 has the fastest accumulation of active ingredients per unit dry weight,and fresh weight accumulation of yu00 is the fastest.The in vitro plantlets of P.polyphylla var.yunnanensis can quickly adapt to the acclimated environment and can accumulate a large amount of active ingredients in a short time.The successful selection of different strains not only helps to compensate for the defects of the germplasm sources in the market,but also provides an excellent source for the market of Paris,and provides an effective,safe and convenient preservation method for the rare Paris species.
