Hainan Qinglangang Mangrove Provincial Nature Reserve(Bamenwan Mangrove forest),located in Wenchang City,Hainan Province,is one of the nature reserves which possess the richest mangrove resources, foster the most vigorous growth of plants, and keep the most intact vegetation communities. The occurrence of main pests was investigated by route survey from July 2017 to December 2018,and the moths diversity was systematically investigated by light trap.Species were identified based both on morphological characters and DNA barcodes.Besides, the community diversity of moths of different months was also analyzed.1.Totally 18 species of pests from 4 orders and 13 families were indentified feeding on mangrove plants during the survey.larva of Corydalis sp.was found feeding on the fruits of Lasiognalha cellifera Meyrick for the first time;Orgyia poslica Walker.Teldenia specca Wilkinson,Trabala vishnou Lefebure were the main pests. The species of defoliators were much more abundant than feeding on fruit and stem.2.A total of 147 species of moths belonging to 19 families were collected by light trap. The checklist of moths from Qinglangang mangroves were established and most of the species were illustrated.Among them,57 species are from the family Noctuidae.accounting for 38.78%of the total species number. The number of species and individuals of the moths collected in different months showed a single peak pattern, with the highest in August and the lowest in January and March.Noctuidae is the dominant family, with the largest number of species in other months.3.Diversity parameters including Shannon-Wiener,Berger-Parker,Margalefd have the similar trend, with a single peak in August. While Pielou is maintained at a high level, with two peaks occurs in May and August.Noctuidae,Geometridae,Limacodidae、Pyralidae,Arctiidae,Lymantriidae and Tortricidae occur in every month, while Cossidae.Lasiocampidae and Sphingidae only occur in June and August;Eupterotidae,Cyclidiidae and Elachistidae only occur in January;Uraniidae only occurs in March.4.DNA barcodes of 125 species of mangrove moths were successfully sequenced-Among them 37 species were sequenced for the first time.A DNA barcodes dataset of mangrove moths in Qinglangang was established.A phylogenetic tree was also reconstructed based on the barcode gene.5.The main structure of the moth phylogenetic tree based the mitochondrial COI gene is as follow:(Noctuoidea+(Geometroidea+Bombycoidea+Gelechioidea+Zygaenidae)+Pyraloidea)+Tortricoidea). The superfamily Noctuoidea and the Gelechioidea which includes Sphingidae,Eupterotidae and Lasiocampidae in this phylogenetic tree are not monophyletic. |
