| In recent years,extreme high-temperature climates around the world have become more frequent.High-temperature disastrous climate has become one of the most important abiotic stress factors affecting crop yields.Extreme high-temperature climate will affect the growth and development of corn.High temperatures can abrogate maize kernels,degrade quality,and ultimately lead to a decline in corn production.The maize backbone inbred line HZS was used as a material to study the differences in gene expression under the influence of high temperature at maize seedling stage by using transcriptome sequencing technology,and to find some maize genes that respond to high-temperature stress,in order to elucidate the molecular mechanisms of maize seedlings’ heat tolerance.The foundation also provides some references for corn breeding research.The main results of the experiment are as follows:1)This experiment used the lllumina HiSeq platform to measure a total of 6 samples of the corn seedling high-temperature treatment group and the control group.The average yield of each sample was 6.11 Gb,and the correlation between the three biological replicates was good.After the clean reads reached 85.8%,the data can be used for further analysis.2)The results of gene expression screening showed that there were 6862 differentially expressed genes in corn seedlings treated with high temperature stress compared with the control group.Among them,3342 differential genes were up-regulated and 3,520 differential genes were down-regulated.3)Analysis of GO function showed that 2985 genes were annotated to biological process classification in differentially expressed genes;3449 genes were annotated to molecular functional classification;3363 genes were annotated to cell component classification.A total of 42 entries were enriched(P-value ≤ 0.05).The most enriched genes in biological processes are classified into transmembrane transport,single biological processes,and photosynthesis;the most enriched genes in molecular function classification are ATP binding,purine nucleoside binding,and purine ribonucleoside binding,etc.The most enriched genes in the cellular components are classified into the thylakoid part,the thylakoid and the inherent components of the membrane.After significant KEGG enrichment analysis,significant pathways for photosynthesis,glucosinolate biosynthesis,and biosynthesis of kerberalin and wax were screened.4)Further in-depth analysis of the annotated results of differentially significant genes,in which SAUR-mediated auxin metabolism,abscisic acid,and gibberellin may be involved in the response regulation of maize seedlings to heat stress;in signaling,the MAPK cascade The response may be involved in the process of high temperature stress response in corn seedlings;MYB,b HLH,AP2-EREBP,WRKY and NAC family of transcription factors may play an important role in maize seedling response to high temperature stress and s HSP20 family and corn seedling response to high temperature stress.The process is closely related... |
