Plant lectins play an important role in defense to disease and insect.Our previous studies showed a rice lectin OsMBLl had role in Pattern-Triggered Immunity(PTI).In this study,we further determined OsMBLl interacting proteins from rice,OsGDSL1 and OsPIP5K,by yeast two hybrid.OsMBL1 were then identified to interacte with OsGDSL1,OsPIP5K N-/C-termius by pull-down immunoprecipitation.The BiFc assay in Nicotiana benthamiana and rice cells both show the interaction of OsGDSL1 and OsMBL1 occured on plasma membrane.OsGDSL1 was encoded with a GDSL-like lipase and had hydrolytic enzyme activity toward lipase.The transient expression in tobacco assay showed that OsGDSL1 was localized onto plasmamembrane.OsGDSL1 wes induced upon exposure to Magnapothe oryzae or plant hormone.The overexpression or konck-out mutants of OsGDSLl were gernerated by agrobacterium-mediated transformation of mature rice embryo.Overexpression of OsGDSL1 in rice conferred resistance against rice blast,in contrast,deletion of this gene compromised resistance against rice blast.In addition,OsGDSL1 transcriptionally regulated OsMBL1 gene.Taking together,this study indentified the interactions beteen OsMBLl and its partener proteins,and preliminarily analyzed fuctions of those genes.These results would provide a theoretical basis for the further of mechanism of rice disease resistance.
