| In this study,inulin was used to prepare novel prebiotic-phthalic anhydride inulin nanoparticles by chemical modification,and Pediococcus acidilacticiy which inhibits intestinal Salmonella in the livestock,was used as a probiotic.Sodium alginate-chitosan-sodium alginate was used as the wall material to prepare microcapsules containing inulin nanoparticles.The test components consisted of four groups.The control group of Pediococcus acidilactici not coated,abbreviated as(PA),the test group I was individually coated with Pediococcus acidilactici,abbreviated as ACA(PA),and the test group Ⅱ was coated with inulin and Pediococcus acidilactici,abbreviated as ACA(PA-I),test group Ⅲcoated inulin nanoparticles and Pediococcus acidilactici,abbreviated as ACA(PA-PIN).The characterization and chemical properties of inulin nanoparticles were studied by advanced analytical methods such as’H-NMR,DLS,and SEM.The gastrointestinal tract tolerance of Pediococcus acidilactici microcapsules with inulin nanoparticles was studied In vi tro.The results showed that inulin nanoparticles with an average diameter of about 200 nm can be prepared by chemical methods.Using the FITC labeling method,it was found that this inulin nanoparticles can be internalized into Pediococcus acidilactici.In addition,the microencapsulation rate of sodium alginate-chitosan-sodium alginate as the wall material can reach 99.99%.The results of test of artificial gastrointestinal tract showed that the survival rate of Pediococcus acidilactici in the ACA(PA-PIN)group was higher than that of the other groups(P<0.05).There was no significant difference in the survival rate of Pediococcus acidilactici between the groups in artificial intestinal fluid(P>0.05).Artificial intestinal fluid quickly released within 30 minutes,and there was no significant difference between the treatment groups(P>0.05).In summary,the inulin nanoparticles produced by chemical method self-assembly can enter the body of Pediococcus acidilactici.The microcapsules containing inulin nanoparticles and Pediococcus acidilactici are more tolerable in artificial gastrointestinal fluid than coated inulin and Pediococcus acidilactici microcapsules and individually coated with Pediococcus acidilactici microcapsules.It is believed that inulin nanoparticles act as a new type of prebiotics and provide scientific basis for the development and utilization of inulin feed resources.More importantly,this inulin nanoparticles is expected to be applied to the development of synbiotic feed additives for the purpose of replacing antibiotics. |
