Radix Salvia miltiorrhiza Bunge,generally called Danshen,is an important Chinese herb which is most effective for treatment of cardiovascular diseases and inflammations.WRKY transcription factor is one of the largest classes of the plant kingdom transcription factor,which contains conserved transcription factor WRKYGQK sequence in their DNA-binding domain.We cloned one WRKY transcription factor named Sm WRKY54(861bp)by molecular biology methods,which’s promoter containing four W-boxes.Tissue expression pattern analysis revealed that Sm WRKY54 highly expressed in the taproots,and then in tender leaves.Elicitation is one of the most common and effective strategies for stimulating secondary metabolite production in plant tissue cultures.Inducing experiment showed that Sm WRKY54 respond by SA、MJ、GA3、YE、ET and suppression by ABA.Subcellular localization results showed that Sm WRKY54 protein localized in the nucleus.The hairy roots of overexpression and inhibition Sm WRKY54 were required by agrobacterium C58C1-mediated transformation of sterile blades.HPLC analysis showed overexpression of Sm WRKY54 could increase the content of tanshinone,but reduced the content of salvianolic acid.We overexpressed Sm WRKY54 in Arabidopsis thaliana and then detected gene expression of five maker genes in SA pathway.The transcriptional levels of those genes were all elevated,which revealed that Sm WRKY54 may be involved in plants’ stress resistance.
