High-speed counter current chromatography is a continuous and efficient liquid-liquid distribution chromatography technology.The instrument based on this technology is called high-speed counter current chromatograph.At present,as an important tool for the separation and purification of substances,the instrument has been widely used in the fields of bio-medicine,chemical and environmental protection.This instrument has a good market prospect.China is one of the first countries to apply high-speed counter current chromatography technology.However,in recent years,due to the lack of in-depth research in the instrument structure innovation design,dynamic performance optimization and precision manufacturing technology,the domestic counter current chromatograph has a certain gap with foreign advanced instruments in product performance,stability and other aspects.Therefore,it is necessary to develop efficient and stable counter current chromatography technology research to realize the independent development of high-efficiency counter current chromatograph,so as to promote the development of domestic separation industry.This paper is supported by the National Natural Science Foundation of China’s major scientific research instrument development project: "Development and application of high efficiency and high speed counter current chromatograph".In order to develop a counter current chromatograph with a centrifugal acceleration of up to 500 g,the structural design,dynamic and static performance analysis,structural topology optimization,whole mode test,vibration test and sample separation experiment of the chromatograph were carried out.The main research work and achievements of this paper are as follows:(1)Based on the separation principle of counter current chromatograph and the design requirements of 500 g centrifugal acceleration,the overall structure design and three-dimensional modeling of the counter current chromatograph with high centrifugal acceleration were carried out.Considering the phase tuning principle of planetary gear,the design criterion of helical gear and the structural characteristics of chromatograph,a set of helical gears with lower internal excitation was designed for the rotor system of chromatograph.(2)The finite element analysis model of the rotor of counter current chromatograph with high centrifugal acceleration was established,and the rotors with different planetary frame materials were analyzed by ANSYS software.The material selection of the planetary frame was completed by comprehensively considering the cost and performance factors.The finite element model of the whole counter current chromatograph with high centrifugal acceleration was established,and the rationality of the structure design was verified by performance analysis.The sample separation experiment was carried out on the high centrifugal acceleration counter current chromatography prototype,and the performance of the prototype was evaluated according to the results.(3)In view of the deficiency of high centrifugal acceleration counter current chromatograph,an improved scheme of chromatograph structure was proposed.The high centrifugal acceleration counter current chromatograph was improved by changing the cantilever structure into a double support structure,replacing the unwinding hose with a metal unwinding frame,and increasing the revolution radius to reduce the rotating speed.Then,the rotor system with improved high centrifugal acceleration counter current chromatograph was optimized by using the optimization software GENESIS.After optimization,the new rotor has slightly improved in terms of dynamic and static performance,but significantly decreased in quality,thus the goal of lightweight design was achieved.(4)The hammering modal test of the improved high centrifugal acceleration counter current chromatography prototype was carried out by using the vibration and analysis platform M + P,and the first 6 order test modal frequencies of the chromatograph prototype were obtained.The consistency between the chromatograph prototype and the chromatograph in the design stage was verified by comparing the test values with the simulation values.The vibration of the prototype in working state was measured.According to the measurement results,the vibration intensity grade of the chromatograph met the requirements of long-term operation under the working speed.Under the condition that the dynamic and static performance of the instrument met the requirements,the separation performance of the improved high centrifugal acceleration counter current chromatograph ws tested by conducting stationary phase retention and sample separation experiments under different working...
