At present,traditional Chinese medicine(TCM)industry is at the crucial period of transformation and upgrading from empirical development towards intelligent production.The fluctuating quality of raw herbs has become an important factor affecting the quality of Chinese medicine products,and the lack of effective quality control methods and online detection methods has seriously hindered the process of Chinese medicine towards internationalization and modernization stage.As a substitute for rhino horn,buffalo horn is the raw material of many Chinese medicine products,and its main medicinal ingredients are various amino acids,the fluctuation of its quality determines the quality of the final herbal product.At present,the establishment of quality control methods for buffalo horn is not yet perfect,so it is urgent to establish a method to distinguish the geographical origin of buffalo horn and the rapid detection method of various amino acid content to ensure the uniformity and stability of quality of different batches of buffalo horn.Near infrared spectroscopy(NIRS),as a high-tech analytical technology,has the characteristics of rapid,real-time,low cost and non-destructive samples.In this paper,NIRS is applied to quality control of buffalo horn,and the application of NIRS combined with chemometric methods in quality control of buffalo horn is discussed from two aspects,and the method applicable to the quality control of buffalo horn is developed to ensure the uniformity and stability of the quality of buffalo horn.The main research contents of this paper include:1.Study on determination method of amino acid content in buffalo hornTaking buffalo horn as the research object,phenyl isothiocyanate(PITC)was used to derivate 16 kinds of amino acids.High performance liquid chromatography(HPLC)was used as the analysis method.Through optimizing chromatographic conditions and methodological investigation,a method was established to simultaneously determine the contents of 16 kinds of amino acids in buffalo horn with theanine as internal standard.The results showed that the method was accurate and reliable,which provided data support for the subsequent study on the determination of amino acid content in buffalo horn by NIR and laid the foundation for further research on quality control of buffalo horn.2.Study on distinguishing method of geographic origin of buffalo hornTaking 147 samples of buffalo horn from three origins,Zhejiang,Sichuan and Yunnan as the research object,and analyzing the NIR spectra using TQ Analyst software,a rapid,simple and non-destructive method of discriminating the geographical origin of buffalo horn samples based on NIR spectra was established by comparing the effects of six spectral pre-processing methods on the discriminative model.The results showed that the combination of NIR spectroscopy and discriminant analysis could effectively discriminate samples of three origins with an accuracy of 98.6%.3.Study on the detection of amino acids in buffalo horn by near infrared spectroscopyThe content of 15 amino acids determined by HPLC was used as reference data,and the reference data was corresponded to NIR one by one.Through a series of model parameters optimization such as spectral pre-processing and selection of spectral range,a rapid,real-time and non-destructive method for the determination of 15 amino acids in buffalo horn was established to provide some theoretical support for the content prediction of 15 amino acids in buffalo horn and lay the foundation for the quality control of buffalo horn.
