Actinidia arguta is a kind of perennial vine which requires regular pruning stems during its fruit cultivation.Most of these stems,except for being used for cuttaging and grafting,would be directly discarded,causing waste of resources.In this experiment,the flavonoids in the woody stems were studied to provide more reference value for making full use of the stems of actinidia arguta in the future,so as to achieve the objective of fully utilizing the resource of actinidia arguta.In this paper,the stems of actinidia arguta were used as raw material to extract and purify flavonoids and prepare tablets.The results are as follows:1.Ultrasonic-assisted extraction method was used to extract the crude flavonoids from the stems and leaves of actinidia arguta,and comparison of the extraction yields revealed that the contents of crude flavonoids extraction from the stems and leaves did not differ greatly.However,in the process of decolorization of raw materials,the amount of reagents required for the leaves is large and the time is long,so stems are selected as the main raw materials.The response surface method was used to optimize the process of the ultrasonic-assisted extraction of the crude flavonoids from the stems of actinidia arguta,and the optimal extraction process of the crude flavonoids from the stems of actinidia arguta was obtained as follows: ultrasonic power was 240 W,ultrasonic time was 6 min,ultrasonic temperature was 70 ℃,ethanol volume fraction was 50%,the ratio of liquid to material was 65 m L/g,and the extraction rate of the crude flavonoids from the stems of actinidia arguta was the highest,which was5.71%.At the same time,the optimal process obtained by response surface method was applied to extract the crude flavonoids from the stems of actinidia arguta in different months(June,August,October),and the contents were in the order of October > August > June.2.The AB-8 macroporous adsorption resin was applied to purify the crude flavonoids extraction from the stems of actinidia arguta.The effects of different factors(concentration of loading solution,p H of loading solution,flow rate of loading solution,flow rate of eluent,etc.)on the adsorption and resolution of AB-8macroporous adsorption resin were investigated to explore the optimal purification conditions.The optimal purification conditions obtained after the tests were as follows: loading solution concentration 0.63 mg/m L,loading solution flow rate 3m L/min,loading solutio p H=5,washing volume 40 m L,eluting(ethanol)concentration 80%,eluting flow rate 3 m L/min and eluting volume 30 m L.Under this condition,the purified flavonoid can be obtained to the maximum extent and the purity of flavonoid is 71.25% by testing.Meanwhile,the constituents of flavonoids in the stems of actinidia arguta were analyzed by HPLC and determined to contain rutin and not quercetin.3.Taking the flavonoids of actinidia arguta stems as the main raw material,adding starch,sodium carboxymethyl cellulose,magnesium stearate and other auxiliary materials to make corresponding tablets to expand the application scope of flavonoids of actinidia arguta stems.Based on the comprehensive score as the investigation index,the best prescription of the tablet was optimized by single factor and orthogonal test.The results were as follows: the addition of flavonoids in the actinidia arguta stems was 38%,the addition of starch was 38%,the addition of sodium carboxymethyl cellulose was 8%,the addition of magnesium stearate was0.50%,and the addition of mannitol was 16%.At the same time,the quality of the best prescription tablets was measured from the aspects of weight difference,hardness,disintegration time,etc.The results showed that the various indexes of the flavonoids tablets of actinidia arguta stems were in line with the relevant provisions.
