Abstract/Summary:
In recent years,nanoparticles based on red blood cell membranes have become a potential nano-drug delivery platform due to their good biocompatibility and immune evasion capabilities.In this work,a mesoporous silica rod(MSNR)composite drug nanocarrier coated with red blood cell membrane(RBCM)is presented,which can be used in the combination therapy of chemotherapeutic and photothermal treatment of SK-BR-3 cells.First,rod-shaped MSNR and RBCM were prepared by the sol-gel method and modified hypotonic lysis method.Then,RBCM was wrapped on the surfaces of the mesoporous silica rods by electrostatic adsorption induction.The coating of RBCM layer can reduce both the premature drug leakage and cytotoxicity.At the same time,it can reduce the probability of complex vectors engulfed by the macrophages.Through multicolor colocalization,we studied the detailed process of internalizing drug carriers in SK-BR-3 cells,as well as the kinetics of drug release.Besides,the compound drug carrier has a good p H-responsive drug release characteristics.By loading DOX and indocyanine green,under the combined chemotherapy and photothermal therapy,the drug carrier shows an excellent tumortargeted therapeutic effect.Therefore,such a kind of drug-loaded RBCM@MSNR have broad application prospects in tumor treatments.
