Inflammatory bowel disease is considered to be related to intestinal flora disorders.At present,clinical treatment is mainly carried out by taking high doses of 5-aminosalicylic acid or steroid hormones daily.The disease cycle is long and recurring,which affects the quality of life of patients and poor drugs.There are many reactions.In order to improve the therapeutic effect of IBD,based on the characteristics of high active oxygen concentration in the inflammation site,this paper adopts intelligent responsive molecular design,nano-characteristic control and targeted molecular modification,and uses particle self-assembly and covalent coupling technology to design and construct a package of Chinese medicine Berberine is a smart amphipathic nanocarrier that introduces into the structure the traditional Chinese medicine notoginseng saponins and astragalus polysaccharides derived from the first-line clinical IBD treatment.This project has completed the preparation of OC-B-BBR nano-microspheres and a series of performance characterizations,rich and detailed animal experiments,and analysis of the bacterial flora of the intestinal microbes of mice,which proves that OC-B-BBR is effective from different angles.The therapeutic effect of inflammatory bowel disease.The successful synthesis of nanocarriers was confirmed by 1H-NMR.With the help of the amphiphilic nature of the nanocarrier to self-assemble into micelles in an aqueous environment,the physical and chemical properties of the micelles were evaluated in detail.The microspheres are evenly distributed;the two kinds of scanning electron microscopy micelles maintain good microsphere morphology and good stability.In the medium of pH=7.4,the micelles have good stability within 5 days,the PDI value fluctuates in a small range of 0.2 to 0.4,and the particle size is almost unchanged.Experiments at the level of living organisms show that the corresponding release performance of nano-microspheres is better,and it is responsive to high ROS.In vivo experiments in mice showed that the IBD disease activity index of the mice administered with OC-B-BBR decreased,the colon status was restored,the spleen index increased,and mucosal damage was reduced;at the histological level,the cell infiltration phenomenon was improved,Goblet cell reduction phenomenon,and abscess phenomenon;the level of inflammatory factors and metabolic levels were evaluated.In all,this study is expected to provide a theoretical basis and solutions for breaking through the bottleneck of the treatment of inflammatory bowel disease,and to provide new ideas for the combination of Chinese and Western medicine.
