Nanodrug delivery system has a wide range of applications in tumor therapy and fluorescence imaging,but there are many problems such as poor stability,short cycle time and drug resistance.Synergistic therapy is an ideal treatment method,which can treat cancer cells through multiple paths,overcome drug resistance and improve the therapeutic effect of drugs.In order to solve the key technical problems such as system stability and multi-drug properties,bovine serum albumin(BSA)coated fluorescent carbon quantum dot(CDs)drug carrier was developed,and a pH sensitive sustained release vision nano-drug delivery system was constructed.The drug release properties of glutamine inhibitor(V9302)and chemotherapy drug doxorubicin(DOX)were studied.In addition,a multi-mode synergistic treatment system was established by combining glutamine inhibitor(V9302)with chemotherapy drug doxorubicin(DOX),showing a highly effective anti-tumor effect.Specific research content includes the following three aspects:(1)The drug delivery system BSA@CDs-DOX was constructed and its drug release performance was studied.Green fluorescent CDs was prepared by rapid microwave method.The particle size of CDs was about 2.5 nm,the surface of CDs was rich in carboxyl group,and the biocompatibility was good.CDs-DOX system was formed by electrostatic action,and then coated with BSA to construct BSA@CDs-DOX drug loading system.The drug loading rate of the system was 72.9%.BSA coating enhanced the stability and pH sensitivity of the system,and the ratio of cumulative release rate increased from 4 to 7.5 when pH = 5.0 to 7.4.Cytotoxicity and scratch test results confirmed that the drug release time extended to 72 h,the proliferation and migration of cells have a more lasting inhibition.In addition,the system enables visual tracking of intracellular drug release and tumor cell inhibition.(2)Based on the cellular metabolic pathway,V9032 can cut off the effect of tumor nutrition.The construction scheme of BSA@CDs-V9302 drug delivery system based on pH response was proposed.The mechanism of drug release and tumor inhibition in the system was revealed,and the inhibition effect on MGC-803 activity of cancer cells was realized.The drug release performance study showed that the encapsulation rate of V9302 was up to60.38%,and the cumulative release rate of V9302 at pH = 5.0 was 7.6 times that at pH = 7.4.The cytotoxicity assay demonstrated the tumor suppressive effect of BSA@CDs-V9302 and demonstrated its ability to monitor and follow it in real time for 48 h.(3)A multi-path synergistic therapy nano-drug delivery system BSA@CDs-V9302/DOX was constructed.The delivery system showed obvious pH sensitivity.The cumulative release rate of DOX and V9302 at pH = 5.0 was 3.2 and 1.57 times of that at pH = 7.0,respectively.Cooperative medical system for MGC-803 cell activity inhibition rate is about 4.2 times that of GES-1 cell rate of collaborative CI value less than 1,that the two drugs in tumor therapy create synergies and multi-way effect on cancer cells,effectively improve the utilization rate of the drug and for collaborative treatment system of nano drug research provides a new strategy and direction.
