Termite balls is a symbiotic fungus(Fibulorhizoctonia sp.)first discovered in termite nests by Japanese scholar Kenji Matsuura.At present,there are few reports on the fermentation of termite balls at home and abroad.Therefore,we have carried out liquid fermentation culture on termite balls,mixed it with Cordyceps militaris,extracted its mixed fermentation products,and continuously optimizedmetabolite adenosine culture.The main results are as follows.(1)The liquid fermentation medium and fermentation conditions for termite balls were optimized.An optimization experiment was carried out on the ratio of the liquid fermentation medium of termite balls.The secondary metabolite adenosine after fermentation was used as an index to optimize the composition of the medium.The orthogonal test design was used to determine the best fermentation culture formula:sucrose40 g/L,yeast powder12 g/L,Mg SO43g/L.Based on determining the medium’s composition,the termite balls liquid fermentation’s culture conditions were also optimized.The optimal combination conditions for adenosine production were screened through single factor and orthogonal experiments:the temperature is 24℃,and the rotation speed is 160 r/min.The inoculum amount is 4%,the p H value is the natural value,and the mycelium’s highest adenosine content is 2.66 mg/g.(2)Determination of adenosine content in Cordyceps militaris in liquid fermentation.The PDA comprehensive liquid medium is used as the basic medium under 24℃,the rotation speed is 160 r/min,the inoculum is 3%,and the p H value is the natural value for the liquid fermentation of Cordyceps militaris.The mycelium is extracted and combined.The content of the mycelium’s highest adenosine content is 4.11 mg/g.(3)Antagonistic experiment of termite balls and Cordyceps militaris.The plate confrontation culture experiment tested whether there is antagonism between the termite fungus and Cordyceps militaris.This experiment shows that there is no antagonism between the two strains,so the two fungi can be used as starter strains for compounding fermentation.(3)Optimization of fermentation conditions by mixed fermentation of termite balls and Cordyceps militaris.Optimizing the ratio of the mixed fermentation medium of termite balls and Cordyceps militaris,using the secondary metabolite adenosine after fermentation as an index,and using orthogonal experiment design to determine the best fermentation medium formula:sucrose30 g/L,peptone 12 g/L,KH2PO43 g/L.On the basis of determining the composition of the medium,the culture conditions were studied,and the optimal combination conditions for adenosine production by mixed fermentation were screened out through single factor and orthogonal experiments:the temperature is 24℃,the rotation speed is160r/min,and the inoculum is 5%,the p H value is the natural value,and the mycelium’s highest adenosine content of mycelium is 6.09 mg/g.In conclusion,the liquid fermentation of termite balls and Cordyceps militaris has a good potential for the development of edible and medical fungi fungi,which is worthy of further research and exploration.
