Abstract/Summary:
Hydroxylated soy phosphatidylethanolamine-Methoxy polyethylene glycols 2000(hydroxylated SPE-MPEG2000)was prepared though oxidation,acylation,amidation,hydrogen peroxide reoxidation and other related methods,with soybean phospholipid and methoxy polyethylene glycols as raw materials in this paper.The structure of the product was confirmed by NMR,IR and other methods.The optimal conditions for preparing SPE-MPEG2000 were m(phospholipid):m(acyl chloride)=3:1,the amount of triethylamine was 3m L,the reaction time was 4 hours,and the reaction temperature was 50℃.The optimal conditions for the hydroxylation process was 4m L of lactic acid,3 m L of hydrogen peroxide,3 hours of reaction time,and 45°C of reaction temperature.The properties of hydroxylated SPE-MPEG2000 were measured.The critical micelle concentration was 5×10-4g/m L,the acid value was 4.13mg KOH/g,and it had certain emulsifying and dispersing power which can be used as a surfactant in related areas.The thin film dispersion method was used to prepare blank liposomes CLs,SSL,calcein-loaded liposomes 1、calcein-loaded liposomes 2、PZQ-CLs,and PZQ-SSL.The morphology,particle size distribution and Zeta potential of blank liposomes CLs and SSL were observed and measured by transmission electron microscope,nano-particle size and Zeta potential analyzer.It was found that the particles of SSL were more complete,the particle size was smaller,and the stability was better.The average encapsulation rate of calcein by liposome 1,liposome 2 PZQ-CLs and PZQ-SSL were 5.83%,11.76%,6.54%and10.69%,respectively.The results have shown that liposomes modified with hydroxylated SPE-MPEG2000 have greatly improved the encapsulation of simulated drugs.
