Abstract/Summary:
As a kind of medicine in breeding,veterinary drugs are widely used in the prevention and treatment of animal diseases.With the increasing demand for animal derived food,food safety problems caused by misuse and abuse veterinary drugs occur frequently.High-throughput detection of trace veterinary drugs in complex biological samples has become a hot spot in the field of analysis.Taking human blood as the research object,novel conjugated microporous polymers(CMP)material was fabricated as solid phase extraction adsorbent.By further combination with high performance liquid chromatography(HPLC)and LC-Q-TOF/MS,a high-throughput,non-targeted detection method for multi-residues of veterinary drugs was established.It can provide important technical support for assessing the risk of dietary exposure,exploring the law of human accumulation and clarifying the pathogenesis of diseases.This paper was divided into four chapters.Chapter 1: The possible sources and accumulation ways of veterinary drugs in human body and the harm of veterinary drugs to human body were described in detail.The existing sample preparation technology and detection methods of veterinary drug residues in complex matrix were also introduced.Chapter 2: Super stable CMP material was synthesized by using 1,3,5-triacetylbenzene and 1,4-diiodobenzene as monomers,cuprous iodide and tetrakis(triphenylphosphine palladium)as catalysts,anhydrous toluene and triethylamine as solvent.The CMP material has been successfully prepared through physical characterization and it also exhibits good physical and chemical properties.Then,the CMP material was used as solid phase extraction adsorbent.By further optimizing the parameters of sample preparation and detection instrument,a CMP-SPE-HPLC method for the determination of 12 veterinary drug residues in human serum was established.These results provide important technical support for the detection of trace veterinary drug residues in complex human samples and the correlation research between residual drugs in food and human health risks.Chapter 3: The mass library for 57 veterinary drugs has been established based on LC-Q-TOF/MS.It contains compound name,molecular formula,retention time and product ion spectra under different collision energies.The library makes the detection of veterinary drug residues achieve a leap forward development from targeted detection to non targeted screening.Then,based on the CMP-SPE-HPLC method established in the previous chapter,by further optimizing the related parameters of veterinary drug extraction and solid phase extraction.A simple method was developed for the screening and identification of multi-class and multi-residues veterinary drugs in human plasma.Under the optimal extraction and detection conditions,the 57 veterinary drugs were well related in the linear range and the recoveries were 63%~120%.The RSD of intra-day and inter-day were 0.6%~12.9% and5.5%~15.6%,which met the detection requirements of multi-class and multi-residues veterinary drugs.Finally,the method was successfully applied to the detection of veterinary drugs residues in human plasma of cancer patients.Metronidazole and melenprogesterone were detected in the test samples.Chapter 4: The full work was summarized.
