| With the development of modern medicine,many new medicines have emerged for treating diseases and saving people,especially after the advent of antibiotics,which has greatly prolonged human life.In the in-depth study of antibiotics,researchers found that antibiotics have certain toxic and side effects,and some of them,therefore,improving the efficacy and reducing the toxic and side effects of drugs on the organism have become the key to disease treatment.Effective ways to solve the above-mentioned problems is to construct a suitable drug carrier to improve the efficacy,and at the same time,to use auxiliary drugs to reduce the toxic and side effects of the main drugs in the treatment of diseases.Resveratrol(Res)extracted from a variety of fruits or plants,because of its oxidative resistance,anti-tumor effect,anti-inflammatory and tissue repair capabilities,has become a research hotspot for anti-cancer,anti-inflammatory and other diseases.The depth of transformation application and research has found that the poor growth of resveratrol limits the efficacy of resveratrol.For this reason,studies on the implantation and optimization of resveratrol drug-loading systems have become resveratrol.Drug-loaded anti-tumor anti-inflammatory diseases are the key to treatment.In this thesis,bacterial cells synthesized by microorganisms are cellulose and resveratrol are used as adjuvant drugs.They are implanted into alternative systems for the treatment of tumors and skin diseases,and the effects of treatment are explored.The content and results of related research are as follows:(1)Synthesis of resveratrol in combination with doxorubicin(Dox)for tumor targeting systems.The bacterial cellulose synthesized by microorganisms is used as a raw material,and the bacterial cellulose nanofibers are controlled by a molecular manipulation technique to prepare nanocrystals with a diameter of 30-50 nm and a length of 150-250 nm.Through theπ-πstacking effect,dopamine was used to replace doxorubicin and resveratrol on the nanocrystals,and resveratrol was implanted together with doxorubicin to treat the tumor drug delivery system.On this basis,the loading and release properties of nanocrystals such as doxorubicin and resveratrol were studied.The results showed that the Dox encapsulation blocked 78.53%.Res encapsulates 37.46%.The in vitro drug release of dual-loaded nanocrystals is p H sensitive.Within 96 h,the release of doxorubicin and resveratrol at p H value of 5.4 was 22.42%and 2.41%higher than that under neutral condition,respectively.Using cell assessment technology,the effects of doxorubicin,resveratrol,and both on normal cells and cancer cells were explored.The results show that:nanocrystals loaded with doxorubicin have a strong killing effect on 4T1cells(mammalian breast cancer cells),and damage to 293T cells(human kidney epithelial cells) is relatively large;nanocrystals loaded with resveratrol are not toxic to 4T1 cells and 293T cells;however,under the synergistic effect of doxorubicin and resveratrol,double drug-loaded micelles has obvious killing effect on 4T1 cells and less toxic effect on 293T cells.It can be shown that the resveratrol combined with adriamycin for the treatment of tumor drug delivery system has obvious antitumor effect in vitro,and also has the function of maintaining normal cell morphology and vitality,and has certain practical potential.(2)Reconstruction of resveratrol in combination with erythromycin(E)in the treatment of skin inflammation through the percutaneous interventional system.β-cyclodextrin was grafted on the bacterial cellulose dissolved in the alkali urea system with epichlorohydrin,and Fe3O4magnetic nanoparticles were added to form a mixed solution,and the mixed solution was added dropwise to the calcium chloride solution to prepare a magnetic gel.An alternative system for resveratrol combined with erythromycin for acne was synthesized.Based on this,the drug release performance of the drug-loaded gel with or without an external magnetic field was compared.A period of 20 minutes was set,and no magnetic field was added during the first 10minutes of the cycle,and a magnetic field was added after the last 10 minutes.The results show that the drug release of the gel is magnetically responsive.In the first cycle,the gel released8.51%of resveratrol in the first 10 minutes(without magnetic field);Magnetic field),the release of resveratrol became significantly faster,and 19.44%of resveratrol was released,which was10.93%more than when no magnetic field was applied.And continuous time increases,within one cycle,the amount of resveratrol released after adding a magnetic field is always greater than that without a magnetic field.Through the bacteriostatic zone experiment,the microorganisms(Staphylococcus aureus,antibiotics,and propionibacterium acnes)on the skin wounds of magnetic gels loaded with erythromycin,resveratrol,and erythromycin and resveratrol compound drugs were evaluated)Antibacterial ability.The results showed that the antibacterial effect of magnetic gels loaded with erythromycin or resveratrol alone was poor,while the antibacterial effects of magnetic gels loaded with erythromycin and resveratrol significantly increased.Magnetic gel loaded with resveratrol and erythromycin to integrate an alternative system with magnetic control function has obvious synergistic effect in the treatment of skin and skin diseases,showing that the drug system has certain effects in the treatment of skin diseases
