Sinomenine is an alkaloid extracted from the dry cane of the deciduous tangled vines,Sinomenium chinense and Caulis Sinomenii.Sinomenine has analgesic,anti-inflammatory,immunosuppressive,and anti-tumor effects.Sinomenine hydrochloride preparations have been used in the clinical treatment of rheumatism,rheumatoid synovial fluid,ankylosing spondylitis and anti-arrhythmia and other diseases,with significant effects.The acid-water extraction method has simple operation process,low energy consumption,simple solvent recovery,and good extraction rate.It is used to extract sinomenine hydrochloride from Caulis Sinomenii.However,the amount of acid and base used in the extraction and purification process is relatively large,and sinomenine is unstable under alkaline conditions,resulting in serious loss of sinomenine hydrochloride and low yield.In addition,there are few studies on the degradation pathway of sinomenine under strong alkaline conditions,and the structure of the degradation products of sinomenine is still unclear.Therefore,this paper studies the extraction and separation process and degradation pathway of sinomenine hydrochloride,optimizes the extraction and separation process,reduces the degradation loss of sinomenine hydrochloride under alkaline conditions,and improves the yield of sinomenine hydrochloride products.Therefore,after analyzing and studying the routes of the original acid water seepage,alkalinization and chloroform extraction,the extraction process parameters are optimized by single-factor experimental method,the amount of acid and alkali is reduced,and the yield of chlorate is improved.To explore the effect of microfiltration/ultrafiltration on the de-noise of the chlorotoxic acid leachate to reduce the effect of impurities on the follow-up process,to study the reaction dynamics of citrine under alkaline conditions,to further identify the degradation products of citrine,and to speculate on the degradation reaction mechanism of citrine,and finally to screen the antioxidants of it to reduce the loss of citrine in alkalinization and improve the recovery rate of it.It was found that the optimal process conditions for the extraction of cyanite-alkali hydrochloride were: The concentration of hydrochloric acid for immersion and percolation is0.1M,and when the p H is adjusted to 11-12 by calcium hydroxide,the effect of alkalization and impurity removal is the best,and the yield of sinomenine hydrochloride is 22.60mg/g medicinal material;Using lime milk,calcium hydroxide and calcium chloride instead of calcium hydroxide for alkalization,the recovery rate of sinomenine was not significantly reduced,and the amount of precipitation was reduced by 44.8% and 42.8% respectively;After the percolate was subjected to microfiltration and ultrafiltration,the solid content was reduced by 10% and 30.2%,respectively,and the sinomenine recovery rates were 93% and 91%,respectively;Sinomenine is prone to oxidative polymerization under alkaline conditions to produce double sinomenine molecules.Adding 0.05% L-cysteine ??can effectively inhibit the oxidative degradation of sinomenine molecules.The research results provide a reference for the research on the production process of sinomenine and the improvement of production efficiency.
