| Liushenqu,also known as Liuqu and Shenqu,is a traditional Chinese medicine koji made by natural fermentation after mixing five kinds of herbs,namely,Polygonum hydropiper,Artemisia annua,Xanthium sibiricum,adzuki bean,bitter almond,wheat bran and flour in a certain proportion.The main purpose of this project is to improve the fermentation method to make the quality of liushenqu more stable.Complete the experimental research on the processing,utilization and extraction of effective components of plants,record the experimental data,optimize the experimental method through the data to evaluate the quality of liushenqu,and reveal the fermentation law of effective components of liushenqu by monitoring the content change of fingerprint substances in the fermentation process,so as to provide theoretical basis for the quality control of liushenqu,And provide a reference for the establishment of quantifiable quality standards in the future,and provide a reference for the future industrial production and the establishment of scientific and effective quality evaluation standards.The main results are as follows.Firstly,the single factor control variable method was used to control the temperature,humidity,fermentation time and the ratio of material to water.After the fermentation,the amylase and proteinase activities were determined by iodine starch colorimetry and Folin phenol method.The results showed that the optimum fermentation conditions were as follows:temperature 30℃,humidity 80%,ratio of material to water 1.5:1,volume of starter 7×7×2(cm×cm×cm)The activity of amylase and protease reached 665.2 U/g and 67.8 U/g respectively.Secondly,through the extraction of effective components from liushenqu and the investigation of chromatographic conditions,the optimal HPLC method for the determination of fermentation fingerprint of liushenqu was determined.The recoveries of three substances were calculated.The absorption wavelength is 360 nm;Methanol was used as the extraction solvent;The mobile phase consisted of methanol-0.2%phosphoric acid solution;The reflux extraction method was selected;The mobile phase consisted of methanol(a)-0.2%phosphoric acid aqueous solution(b).The elution gradient was 0-30min,35%-65%(A)。The contents of quercitrin,quercetin and Luteolin in liushenqu were determined by HPLC.The contents of the three components were 15.4%by external standard method μg/mL、5.2 μg/mL、8.6 μg/mL。Thirdly,16S rRNA was used to analyze and identify the bacteria in the microbial community of liushenqu,and the self-made liushenqu was obtained by the flora grade abundance diagram,and the flora content was rich and uniform;The results showed that the number of bacteria increased in the late stage of fermentation;According to the species abundance heat map,digestive system and endocrine system are metabolic pathways related to drug function in the early stage of fermentation,which are more active in the early stage of fermentation;In the early stage of fermentation,Enterobacteriaceae accounted for 90%.Finally,the industrial production of 50kg was carried out,and the transition from small-scale laboratory production to large-scale industrial production was initially completed.In order to achieve large-scale and standardized mass production of high-quality liushenqu preparation,the annual output of 100 t liushenqu patent medicine was initially realized,so as to ensure the production quality and medication safety of the product.By adjusting the industrial process parameters,optimizing the industrial production process and determining the industrial fermentation production process of liushenqu,the appearance and the detection of effective components meet the standards,realizing the transition from laboratory production to industrial production. |
