| Liver injury is the common pathological basis of many liver diseases in clinic,and continuous injury may eventually lead to liver failure.Currently,the treatment of liver injury is mainly based on western medicine.Western medicine has made some progress in the treatment of liver damage,but there are still some side effects.Longdan Xiegan preparations is a traditional Chinese herbal compound,which is mainly used to treat liver inflammation.The prescription of traditional herbal medicine in our country is a synergistic combination of a variety of herbal medicines,advocating joint medicine and integrity.According to the theory of traditional Chinese medicine,traditional Chinese medicine prescriptions treat diseases in the form of synergistic pharmacological effects of the active ingredients of each traditional Chinese medicine.In this study,by means of gas chromatography-mass spectrometry(GC-MS)and metabolomics methods,the protective effect and mechanism of Longdan Xiegan preparation on liver injury were studied.The results showed that Longdan Xiegan Decoction had a protective effect on acute liver injury by regulating 8 metabolic biomarkers,including pyruvate,sarcosine,fumaric acid,succinic acid,L-allothreonine,4-hydroxyproline,citric acid and indole lactic acid.These biomarkers were related to 5 metabolic pathways,including citrate cycle(TCA cycle),glycine,serine and threonine metabolism,glyoxylic acid and dicarboxylic acid metabolism,pyruvate metabolism and glycolysis or gluconeogenesis.Longdan Xiegan Granules had a protective effect on oxidative liver injury.Its mechanism was related to regulate 18 metabolic biomarkers of liver homogenate and blood.These biomarkers belonged to 11 metabolic pathways,which were arginine biosynthesis,alanine,glutamic acid,aspartic acid and metabolism of phenylalanine,tyrosine and tryptophan biosynthesis,TCA cycle and tyrosine metabolism,D-glutamic acid and D-glutamine metabolism,metabolism of butyric acid,arginine and proline metabolism of phenylalanine metabolism,glyoxylic acid and dicarboxylic acid metabolism,ubiquinone and other terpene quinone biosynthesis.In addition,it was also regulate20 metabolic biomarkers of urine.These biomarkers belonged to 11 metabolic pathways,which were Alanine,aspartate and glutamate metabolism,phenylalanine,tyrosine and tryptophan biosynthesis,TCA cycle,glyoxylate and dicarboxylate metabolism,phenylalanine metabolism,glycine,serine and threonine metabolism,butanoate metabolism,purine metabolism,ubiquinone and other terpenoid-quinone biosynthesis.The results also showed that gentiopicroside had a protective effect on Alcoholic liver injury(ALD),and the mechanism was related to the regulation of 88 biomarkers and 19 metabolic pathways.The main pathways were the TCA cycle,mitochondrial electron transport chain(glycerophosphate shuttle),pentose phosphate pathway,glycerolipid metabolism and glutathione metabolism.Furthermore,gentiopicroside also had antiapoptosis effect.In conclusion,Longdan Xiegan preparation has a certain protective effect on liver injury in rats.The experimental results of this paper can provide preliminary experimental data for the further development of Longdan Xiegan preparation as a liver-protecting drug. |
