| Compared with other countries,my country’s yak blood resources are relatively rich,most of which are treated as slaughterhouse waste,causing serious environmental pollution and a huge waste of resources.In order to realize the industrial production of slaughter waste blood amino acid liquid fertilizer,this paper uses yak blood as raw material,based on the optimization of raw material pretreatment methods such as anticoagulants,protein denaturants,and lye adjustment methods,and establishes a composite based on the optimization of a single enzyme hydrolysis process.Enzymatic step-by-step hydrolysis process to identify peptides,amino acids,elemental composition and content in the hydrolysate,prepare yak blood amino acid liquid fertilizer through blending;and grow Shanghai greens and spinach through greenhouses to promote growth,increase yield,and increase agricultural products The effect of yak blood amino acid liquid fertilizer was evaluated in three aspects of quality,and the following research results were obtained:1.The pre-treatment study found that 1-3%sodium citrate and 1%low concentration ammonium oxalate can improve the rate of yak blood proteolysis,but it is not significantly different from the control group.High concentration EDTA has an inhibitory effect on enzymatic hydrolysis;6 mol/L urea and 3 mol/L guanidine hydrochloride have a better effect on promoting the hydrolysis of alkaline protease,and guanidine hydrochloride has an inhibitory effect on flavor protease.The physical high temperature denaturation at 75-85℃has no significant effect on the hydrolysis rate of the two enzymes;the results of the hydrolysis rate of fresh blood and blood meal are not significant,so blood meal can be used for follow-up research;in addition to Na2HPO4,Na OH,Ca(OH)2and KOH lye Both can be used to adjust the p H of yak blood;before the enzymatic hydrolysis of yak blood,there is no need to perform anticoagulation,protein denaturation and other treatments,and only need to adjust its p H with lye to maintain better conditions for enzymatic hydrolysis.2.In the optimization of yak blood enzymatic hydrolysis conditions,the hydrolysis effects of 8 common proteases on yak blood were compared and analyzed.The hydrolysis effect of flavor protease was the best,followed by alkaline protease;single factor experiment and five factor three level orthogonal experiment were adopted.The process conditions for the hydrolysis of yak blood by flavour protease and alkaline protease were optimized:single factor experiment combined with orthogonal experiment to obtain the optimal hydrolysis parameters of alkaline protease as material-liquid ratio 1:6 g/m L,enzyme concentration 1.25%,optimum p H9,The optimum temperature is 55℃,the hydrolysis time is 6 h,and the protein hydrolysis rate is 13.56±0.39%;the optimum hydrolysis parameters of flavor protease are 1:9 g/m L,enzyme concentration 1.25%,optimum p H 6,optimum temperature 50℃,hydrolysis time is 6 h,protein hydrolysis rate is 33.42±0.44%.Finally,it was determined that the process parameters of the compound enzyme stepwise hydrolysis were as follows:material-to-liquid ratio 1:6 g/m L,p H 9,hydrolysis temperature 55℃,1.00%alkaline protease hydrolyzed for 1 h,adjusted p H to 7,added 0.75%flavour protease to hydrolyze for 5 h,yak The hydrolysis rate of blood protein was 30.77±0.18%.3.The analysis of the amino acid,peptide and element results of the hydrolyzed product found that the enzymatic hydrolysis of yak blood increased the content of amino acids in the hydrolysate.The peptides in the complex enzyme hydrolysate were mainly concentrated in 7-9 peptides,and the limited elements did not exceed the standard.The sensory evaluation of yak blood amino acid liquid fertilizer is better,and the physical and chemical indicators meet the relevant standards.4.Explore the effects of yak blood amino acid liquid fertilizer on crops from three aspects:growth promotion,yield increase,and quality improvement.In terms of growth,BYAAF group has a significant effect in promoting stem thickness,and the effect of spinach is better than that of Shanghai green;in terms of increasing yield,the AAF group has the best effect;in terms of quality,the YBAAF group has increased some active substances in the crop,but the nitrate content is the highest among the five treatment groups,Vitamin C content is also higher than other groups.Generally speaking,the fertilization group is better than the control group,and the organic fertilizer is better than the chemical fertilizer group.Among the organic fertilizer groups,the commercial amino acid liquid fertilizer is the best,and the yak blood enzymatic amino acid fertilizer is slightly lower,But there was no significant difference with farm manure. |
