Metal-organic frameworks(MOFs)are a new type of hybrid porous materials bridged by metal ions and organic groups,which have attracted more and more attention in recent years.The excellent properties of MOFs: definite pore size,adjustable composition and structure,adjustable size,high drug loading and good biocompatibility,make it have a good application prospect in the field of biomedicine.Zeolite imidazole ester framework materials(ZIFs)are one of the MOFs materials.In this thesis,the zeolite imidazole ester framework material ZIF-90 encapsulated flavonoids(chrysin and naringenin),synthesized three different ZIF-90 materials,and further systematically studied these three ZIF-90 materials Interaction with DNA and protein.This thesis mainly expands from the following six parts:The first chapter systematically summarizes the preparation and application of zeolite imidazole ester framework materials;introduces several common biological macromolecule models(DNA and protein);summarizes the research methods of the interaction between drugs and biological macromolecules;finally,proposes The thought and research significance of this article’s topic selection are discussed.Chapter 2,using the one-pot method,using ZIF-90 material to encapsulate chrysin and naringenin,three ZIF-90 materials were obtained,namely the unencapsulated ZIF-90 and the Chr@ZIF-90 that encapsulated the chrysin and Nar@ZIF-90 encapsulating naringenin,and further use powder X-ray diffraction(XRD),Fourier infrared spectroscopy(FTIR),transmission electron microscope(TEM),X-ray photoelectron spectroscopy(XPS)and thermogravimetric(TGA)methods are used to characterize these three ZIF-90 materials.The results showed that both chrysin and naringenin were successfully encapsulated in ZIF-90.Chapter 3,using ultraviolet-visible absorption spectroscopy and fluorescence spectroscopy,studied the mechanism of action of three ZIF-90 materials with calf thymus DNA(ct DNA);using electrochemical methods and molecular docking,studied three ZIF-90 materials The main force with ct DNA.The results show that there is no obvious difference in the mechanism and main force between the three ZIF-90 materials and ct DNA.Chapter 4,using fluorescence spectroscopy and ultraviolet-visible absorption spectroscopy,studied the quenching mechanism and main effects of three ZIF-90 materials on the endogenous fluorescence of bovine serum albumin(BSA);using site competition and molecular docking,The effect sites of three ZIF-90 materials on BSA were studied;synchronous fluorescence,three-dimensional fluorescence and Fourier infrared spectroscopy were used to study the effects of three ZIF-90 materials on the secondary structure of BSA.The results show that the three ZIF-90 materials have different sites of action on BSA,further indicating that both chrysin and naringenin are successfully encapsulated in ZIF-90.Chapter 5,using fluorescence spectroscopy and ultraviolet-visible absorption spectroscopy,studied the quenching mechanism and main force of three ZIF-90 materials on the endogenous fluorescence of human serum albumin(HSA);using synchronous fluorescence,three-dimensional fluorescence and Fu Liye infrared spectroscopy,studied the effects of three ZIF-90 materials on the secondary structure of HSA.The results show that the quenching ability of the three ZIF-90 materials to HSA’s endogenous fluorescence is different.Chapter 6,summarizes and looks forward to the full text.
