The rapid development of agriculture has led to an increase in the types of pesticides and their use more and more widely.The problems of various pesticide residue are becoming more and more serious.In recent years,the detection methods for pesticide residues have also been fully developed.Among them,the detection technology of pesticide residues based on nucleic acid aptamers plays an important role in daily life.Compared with antibodies used in traditional detection methods,aptamers are easier to synthesize and modify,and there are more types of targets that can be detected by aptamers.In recent years,some research methods have combined aptamer with fluorescence detection,electrochemical detection,Raman spectroscopy and other methods.These methods can detect pesticide residues more accurately and sensitively.Some of the new methods also achieve high-throughput detection.However,the detection time of these methods is relatively long and does not meet the high-throughput requirements of daily detection.Therefore,it is necessary to develop a high-throughput,low-time-consuming method.In this research,we first have a good grasp of the floating chip technology,and design the corresponding experimental principles.Five aptamers for pesticide residues were determined,and new complementary chain fragments were designed and verified.The microspheres with different numbers are used for qualitative analysis,and SA-PE that can be coupled with biotin on the complementary chain is used for quantitative analysis.A high-throughput,high-sensitivity,and rapid detection platform for estradiol,bisphenol A,polychlorinated biphenyls,atrazine,and profenofos was constructed.The aptamer used in this platform has strong binding ability to small pesticide residue molecules,and the newly designed complementary chain fragments can improve the sensitivity of the method compared with the commonly used ones in the past,and shorten the detection time,and the detection can be completed in about 40 min.On the basis of this platform,a high-throughput rapid detection kit for the detection of pesticide residues was designed and verified.The kit can be stored for about 20 days at 4 °C,and it is best to use within 7 days.The final results show that the sensitivity of this method is improved compared with other methods,and the minimum detection limits for atrazine,profenofos,bisphenol A,estradiol,and PCBs are 9.4653,20.7514,23.8094,8.9712,6.2677 pg/m L.The recoveries of the diluted pesticide residues spiked in the Haihe samples ranged from 87.5% to 111.02%,and the relative standard deviation(RSD)was less than 28.44%.This method and kit provide at least dozens of combinations for high-throughput detection of pesticide residues,and have broad prospects and development in high-throughput rapid detection of pesticide residues. |
