| Carbon has a valence of+2 in carbon monoxide,and can be further oxidized to+4,which makes it combustible and reductive.From the discovery of CO to its research process,There is no doubt that the development of CO is tortuous.At present,it has been proved that carbon monoxide known as"silent killer",is not only a harmful gas to animals and plants,but also an extremely important gaseous transmitter molecule.Although the amount of CO is small in the body of life,it plays an important role in normal pathology and physiology.The change of carbon monoxide level will cause some diseases.The monitoring of CO has research value and significance in the body of life,so it is urgent to find a convenient and rapid method to monitor the change of CO.In this thesis,three fluorescent probes of CO were designed by using nitro reduction and Tsuji-Trost reaction.The main features include the following aspects:(1)Based on naphthalimide and 2-(2-aminoethyl)pyridine,a lysosomal targeting fluorescent probe Na Ly-CO was constructed.The probe Na Ly-CO has many outstanding characteristics,with a detection limit of(79 n M).In addition,it also has good biocompatibility.It can be used to detect CO in complex cell environment and mainly concentrated on lysosome.This is the first time to detect carbon monoxide in lysosome based on 2-(2-aminoethyl)pyridine.(2)Through naphthalimide as the fluorescence skeleton,sulfonamide as the Golgiosome target group,and through the nitro group without metal also originally identified CO,and a Golgi targeted fluorescent probe Na GA-CO was designed.The probe Na GA-CO not only has the characteristics of quick response CO,and adding 200μM CO will be 21.3 times the fluorescence enhancement,and in 0~1.0μM CORM-2 within the scope of probe Na GA-CO detection limit is 368 n M.In addition,cell imaging results show that the probe Na GA-CO has a complex environment detection ability of CO in cells,and the main enrichment on the golgi apparatus.This is the first of fluorescent probe which can detect CO in Golgi body.(3)The carbon fluorescence probe Na-CO,by construction of the first fluorescence quenched monooxide based on naphthalimide.The probe through pd~0 mediated Tsuji-Trost reaction.Its detection limit is 156 n M,and has a high sensitivity.After adding 200μM CO the fluorescence attenuation of the probe is6%.Even under the irradiation of ultraviolet lamp for 99 minutes,the fluorescence of the test solution remains stable,this results indicated that the probe has good stability. |
