Streptomyces,as an important source of drug leading compounds,especially antibiotics.It has attracted much attention.Based on genome mining technology,this paper mined the genomic information of Streptomyces sp.YINM00001,analyzed the metabolic potential of the strains,used different media to culture and systematically studied secondary metabolites.On this basis,the differences of secondary metabolites and their activities in different media were analyzed and compared.Genome mining technology opens a new way for the discovery of natural products and speeds up the research progress of secondary metabolites of Streptomyces.In chapter one,the research progress of genomic mining of secondary metabolites of Streptomyces was reviewed.In chapter two,the genome of Streptomyces sp.YINM00001 was mined,and it was found that the genome size of the strain was 8.69 Mb,and the GC content was 71.63%,which contained 52 biosynthetic gene clusters.The prediction of the biosynthetic gene clusters of the strain had great potential to produce new compounds.Used OSMAC strategy,10 media were used to screen the strain.It was found that the strain produced some different secondary metabolites when cultured on the 4# and 9# media,and the amount of metabolism was relatively large compared to other media.In chapter three,the secondary metabolites of Streptomyces sp.YINM00001 were systematically studied.The fermentation of the strain was expanded on 4# and 9# media.19 compounds were isolated under 4# medium.Including 3-methythioacrylic acid(1),non-3-ene-2,7-dione(2),indole-3-carboxylic acid(3),indole-3-acetic acid(4),prolipyrone A(5),nonactic acid(6),homonon actic(7),bonactin B(8),bonactin(9),three trilactones(10),dinactin(11),actiphenol(12),3-hydroxy-3-[2-(2-hydroxy-3,5-dimethylphenyl)-2-oxoethyl] glutarimide(13),anthracimycin(14),iso-cycloheximide(15),cycloheximide(16),4-[(2-[(3R,5S)-3,5-dimethyl-2-oxocyclohexylidene]ethyl]-2,6-piperidinedione(17),4-[2-[(3S,5S)-3,5-dimethyl-2-oxocyclohexylidene]ethyl]-2,6-piperidinedione(18),naramycin B(19).Compounds 2 and 8 were new compounds.11 compounds were isolated under 9# medium.Including pyrocatechol(20),2,3-dihydroxybenzoic acid(21),pyrrole-2-carboxylic acid(22),2-isopropyl-5,6-dihydro-4Hpyrrolo[1,2-b] pyrazole(23),1,3,5-trimethoxy-7-methylnaphthalene(24),brevianamide F(25),cyclo-(val-pro)(26),valrrnlaotam(27),N-acetyl-tyramine(28),4,8-dimethoxy-6-methylnaphthalen-2-ol(29),3,5-dimethoxy-7-methylnaphthalen-2-ol(30).Compound29 was a new compound.In addition,the cytotoxic activity and NO inhibitory activity of some compounds were evaluated in vitro.On this basis,the types and activities of secondary metabolites under different medium conditions were further compared and analyzed by HPLC in this study,which provided theoretical support for further study of Streptomyces sp.YINM00001. |
