According to statistics,the death toll of cerebral infarction in 2019 reached up to an astonishing two million,and the incidence rate has been rising among younger individuals.As people are getting more aware of their health problems,safe drugs with high effectiveness are urgently needed.Urokinase derived from human body is a natural thrombolytic medicine.It is highly recognized for its clear mechanism of action and natural advantages it possesses to work well in the metabolic process in human body with lower toxicity and fewer side effects.The market demand for human urokinase is huge yet hardly satisfied,because fresh human urine still remains a scarce recourse.The protein in the urine of healthy human is rich in variety.Clinical effectiveness of target protein(urokinase)found in human urine has been proven,but the extremely low concentration makes it difficult to perform enrichment.In order to mitigate the affect of the difficulty in collecting raw material(fresh human urine)on the output of urine protein,this paper presents how higher efficiency in the production of urine protein can be achieved by exploring the quality control of fresh human urine and modifying the parameters of production process in traditional methods.The main contents of the paper are as follows:a.how high product quality can be ensured at the source by controlling the quality of the raw urineIn this paper,the differences on molecular structures and concentrations of protein in the human urine samples from individuals of different age groups,the time interval of urine collection,conditions of storage,transportation and production,challenge from microorganism and other factors that may affect the quality of the urine protein produced are studied.This paper also defines requirements for manufacturers of crude urine protein and gives suggestions that raw urine is suggested to be stored at under 10℃ and put into protein production within 8 hours after being collected before it starts to deteriorate.The urine collected from healthy male human beings should be stored at low temperature.The shorter the duration of storage and transportation,the better.Raw urine collection is suggested to be performed twice every 24 hours to ensure its freshness.And it is necessary to make sure that the collection equipment is thoroughly cleaned to reduce the breeding rate of microorganisms.b.screening of extraction methods of urokinaseBy comparing different types of adsorption media and different sequential arrangements of addition,we are able to test out their adsorption capacities on target protein.And multiple rounds of trying and comparison among different ways of adsorption and desorption were conducted before the adsorption method was found suited for the enterprise production.c.modifications on manufacturing parameters in the process of enrichment and capture of target protein using adsorption method for higher efficiency and yieldOn the basis of the traditional idea of urine protein production,which is to collect urine first and then perform urine protein adsorption in a massive manner,by adjusting certain production parameters,we can achieve continuous output of protein so as to ensure that the amount of the crude urine protein product extracted via the new adsorption process can meet more most of the market demand.In this paper,the following parameters of urine protein manufacturing is confirmed:The grade of silica gel to be applied is suggested to be 80 mesh.The proper concentration of silica gel in urine is 6 liters/ton.Stir the solution for 120 minutes and 70%of the total protein contained can be obtained.These modifications on the parameters make it more convenient to operate for batch production.In this way,the quality of the whole production process can be effectively overseen and better manufacturing compliance can be achieved.The operational eficiency can be improved by 2-3 times,and production is expected to be doubled.In general,the exploration of the adsorption method of urokinase helps satisfy the demand of rapid extraction of target protein.The new adsorption method in the production of crude urokinase products can greatly reduce the labor intensity and duration as well as simplify the operation.By applying the batch production mode under which adsorption media is to be fed in and then fully retrieved after a set time,the workers’ operational intention and the feasibility of the operation itself will both be significantly improved.
