Abstract/Summary:
Curcumin has low blood concentration and low bioavailability due to its poor water solubility and easy precipitation and crystallization.The water solubility of curcumin has become the biggest problem in the research and development of functional food and clinical drugs.In this paper,the preparation conditions and stability of Cur-CS-NPs by ultrasonic treatment were studied,and the inhibitory activity and cell transport characteristics of curcumin casein nanoparticles dispersion on arthritis cells were investigated.(1)The effect of ultrasonic treatment at different time on the physicochemical properties of CS-NPs was determined.After ultrasonic treatment of casein solution,the physicochemical properties and emulsifying properties of casein were significantly changed,and CS-NPs could be prepared.Ultrasonic treatment significantly reduced the particle size,turbidity,surface hydrophobicity,apparent viscosity and surface free sulfhydryl group of CS-NPs,and improved the emulsification of CS-NPs.Compared with 5 min ultrasonic treatment,longer ultrasonic time had no significant effect on the above indexes.Therefore,ultrasonic treatment of casein solution for 5 min may make CS-NPs more stable.(2)On the basis of the preparation of CS-NPs,the casein solution was pretreated by ultrasound for5 min.curcumin / casein 1:10,1.5:10 and 2:10 curcumin were added for different time to prepare Cur-CS-NPs The stability of Cur-CS-NPs was also studied.The results showed that Cur-CS-NPs with10 mg / ml casein solution and curcumin loaded with 1:10 curcumin for 3 min were the most stable.(3)Cur-CS-NPs and curcumin solution were prepared by ultrasonic treatment.The inhibitory activity of Cur-CS-NPs and curcumin solution were studied.It was found that the anti-inflammatory activity of Cur-CS-NPs prepared by ultrasonic treatment was significantly higher than that of curcumin solution prepared by traditional cosolvent.The inhibition rate of Cur-CS-NPs on arthritis was more than90%.The stability of Cur-CS-NPs was improved to a certain extent,and its inhibitory activity on arthritis cells was improved.(4)Cur-CS-NPs significantly reduced the concentration threshold of curcumin on Caco-2 cells.Compared with the curcumin solution prepared by traditional cosolvent(PEG 300 and Tween 80),the apparent permeability of Cur-CS-NPs in Caco-2 cell absorption and transport model was significantly improved,and the bioavailability of curcumin was improved at a certain elevation.In this paper,a theoretical basis for the preparation of drug loaded nano dispersion system by ultrasonic treatment was established.
