Mai-luo-ning(MLN)is the main production product of Jinling Pharmaceutical Co.,Ltd.,and its sales profit accounts for 80%of the company’s profit,but the environmental protection investment for processing production is about 5 million yuan per year.In the production process,MLN waste composition includes residue of dregs,production wastewater and alcohol precipitate.The experiments were aimed at enriching the active small molecular substances in the wastewater of MLN,reducing the cost of wastewater treatment,and obtaining the separated and purified polysaccharide from the alcohol precipitation.In this way,not only the waste of Chinese medicine production can be converted into usable by-products and the cost of environmental treatment can be reduced,but also it can provide a reference model for the resource utilization of the Chinese medicine industry.The research content of this thesis is divided into the following four parts:Experiment Ⅰ Detection of small molecular substances in the wastewater of MLN productionThe active small molecular substances detectable in the wastewater of MLN were mainly chlorogenic acid and caffeic acid,and the content of chlorogenic acid is higher than content of caffeic acid.In this esperiment,the interfering substances in the wastewater,the polymers,were also tested.Through the detection of the content of polymers,it was found that the content of tannin in wastewater was low,while the content of macromolecules such as pectin,starch and protein were high,which may cause membrane fouling on the surface of hollow fiber membrane.The enrichment efficiency of the acid makes it necessary to examine the process of the membrane technology to form an optimal filtration procedure.Experiment Ⅱ Membrane technology optimizes active constituents in enriched wastewaterIn this experiment,hollow fiber membranes with molecular weight of 4 kDa,6 kDa and 10 kDa were used to purify the wastewater from Mailuoning injection.Based on the chlorogenic acid permeability and the common polymer rejection rate,experiments have shown that the membrane model with molecular weight cutoff of 4 kDa has the best retention effect.At the same time,it can be seen from the experimental results that the membrane ultrafiltration process has an excellent retention effect on the common polymer.Experiment Ⅲ Isolation,purification and structure identification of MLN polysaccharideIn this chapter,the structure of MLN-1-1 was analyzed by HPLC-ELSD,GC-MS,periodate oxidation,methylation reaction,FT-IR and NMR.In this study,the structure of the homologous polysaccharide of Mailuoning was studied.The results showed that MLN-1-1 is a neutral polysaccharide similar to the structure of stachyose.Its molecular weight is about 3 220 Da,and its structure may be α-D-Gal-(6→1)-α-D-Glc-α-D-Gal(4→1)is the main chain,and its branch is α-D-Glc-(1→2)-β-D-Fru And repeat 5-9 units of a polysaccharide.Experiment Ⅳ Antioxidant activity of MLN polysaccharide in vitroIn this chapter,the antioxidant capacity of Mailuoning homopolysaccharide was studied.The results showed that MLN-1-1 has certain antioxidant capacity,and its DPPH free radical scavenging ability reached 60.83%at 1.0 mg/mL.The maximum scavenging rates of oxyanion radicals and hydroxyl radicals were 48.2%and 55.3%,respectively,in this test.Thus,MLN-1-1 has the potential to develop into a natural antioxidant.
