Tube cistanche deserticola has broad medical value,the system research about tube flowers c.deserticola is still less,using benzene ethanol ultrasonic extraction tube flowers c.deserticola total glycosides(PhGs),and by using the response surface,process optimization,to optimize the process of pilot scale enlarge validation,use decoloring with activated carbon to the PhGs crude extraction,explore the PhGs decoloring process;Further,macroporous resin was used for separation and purification to conduct preliminary biological activity research on the obtained PhGs,and the PhGs were made into sustained release tablets as the main component.The research results of this paper are as follows:1.Extraction technology of PhGsUltrasonic extraction method was used to extract PhGs from Cistanche tubulosa,and the optimal single factor extraction process was obtained.Response surface was used to optimize the extraction process of PhGs,and the optimal process conditions were obtained as follows: ethanol concentration 70%,ultrasonic power 160 W,ultrasonic time 5 min,liquid to material ratio 1:16.2.Research on PhGs decolorization technologyCrude extractings from color deeper PhGs decoloring with activated carbon,and the bleaching time,bleaching temperature and carbon content in three factors of single factor experiment,the decoloring process were optimized by response surface to get the best PhGs decoloring process condition is: the activated carbon content is 1.50%,53 min bleaching time,bleaching temperature 64 ℃,the PhGs crude extract significantly lighter color,decoloring rate and retention rate to achieve the optimal.3.Study on purification process of PhGsUsing macroporous resin separation and purification of crude extractings from PhGs,through screening and purification of macroporous resin adsorption analytical experiment,then the rate on the concentration of crude extract on sample,column,concentration of eluent,eventually the PhGs purification process for the selection of the sample concentration of 2.5 mg/mL,column on the sample volume 10 BV,70% ethanol aqueous solution as the eluent,will concentrate the PhGs lyophilization of solid powder content is 53.15%.4.PhGs bioactivity evaluationBgc-823 cells and Hacat cells were treated with PhGs at different concentrations to observe the morphological changes of the nucleus.The results showed that PhGs had no toxicity on the cells.The determination of the reduction ability and DPPH scavenging ability of PhGs crude extract from Cistanche tubulosa proved that PhGs substances had certain reduction ability and scavenging ability of free radicals.5.Development of PhGs skeleton sustained release tabletsBy studying the sustained release preparation technology of PhGs,the optimum conditions for the production of sustained release tablets were as follows: 23.0% h PMC-10 W,11.0% lactose and 10.0% magnesium stearate.In this paper,a systematic development process of Cistanche tubulosa was obtained,including the extraction,decolorization,purification and biological activity analysis of the functional components of PhGs,and the exploration of PhGs sustained release tablets was carried out,which provided certain experimental basis for the comprehensive development and research of PhGs,with important application value.
