| Glycosmis pentaphylla,which belongs to the Glycosmis genus of Rutaceae family,is a traditional medicine in the Dai ethnic group of China.It was found that alkaloids isolated from the roots,stems,fruits and other parts of G.pentaphylla had good biological activities.However,the research on the leaves of Glycosmis pentaphylla was rarely reported.In this work,a systematic chemical constituents research on the leaves of Glycosmis pentaphylla have been completed,and the anti-inflammatory and anti-tumor activities of the sulfur-containing amides from the leaves of G.pentaphylla have been evaluated.Forty compounds(1-40)were isolated from the leaves of G.pentaphylla by modern separation technology,including 27sulfur-containing amides(1-27),3 alkaloids(28-30),4 lignans(31-34),2steroids(35-36),2 terpenes(37-38)and 2 phenols(39-40).Among these compounds,compound 4-27 are new sulfur-containing amides,and compound 31 is an undescribed neolignan.In addition,the possible biosynthesis pathways of three special sulfoamides(22,23,27)with six-number ring were also proposed.Sulfur-containing amides(1-27)were assessed in terms of their cytotoxic effects against human tumor cell lines(SGC7901,Hep G-2,MCF-7,T98 and HCT-116).The results showed that most of the sulfur-containing amides had different degrees of inhibition on tumor cells.Especially,compound 2 gave the most powerful inhibition to SGC7901 which was 1.7times higher than that of positive control cisplatin.And compound 16 gave the most powerful inhibition to MCF-7 and T98 were which were 2.6 and 2.9times higher than that of cisplatin,respectively.In addition,the anti-proliferative activities of sulfur-containing amides against Hep G-2were taken as examples to discuss the structure-activity relationships preliminarily.RAW 264.7 cells induced by LPS was used as inflammatory model,and the productions of NO in culture supernatant were measured by Griess method.Compounds 1-3,5-8,18-19,25 exhibited good anti-inflammatory activity,with IC50values of 0.16-16.74μM.It was worth mentioning that compound 19 had the strongest anti-inflammatory activity,which was 60times higher than positive control dexamethasone.Furthermore,the structure-activity relationships of sulfur-containing amides on the anti-inflammatory activities were preliminarily discussed.The results showed that there were various types of compounds in the leaves of G.pentaphylla,including many sulfur-containing amides.Some of these sulfur-containing amides showed pronounced anti-inflammatory and anti-tumor activities,which indicated sulfur-containing amides had the potential research value in the development of new anti-inflammatory and anti-tumor drugs. |
