Bioaerosol refers to aerosol particles containing biologically active components which particle size is between 0.01 and 100 μm,often including microbial particles such as bacteria, fungi and viruses, and particulate components released by organisms such as spores, plant pollen, and animal fragments.Inhalation of bioaerosols can lead to a variety of diseases, posing a potential hazard to human health. At present, there are few related studies on bioaerosol composition analysis, and there is no technology for effectively monitoring the content of different biological components in aerosols. In this study, bioaerosol samples were collected using the BIO-Capturer-6 aerosol collector, and the collected samples were detected using animal, plant, and bacterial housekeeping gene fluorescent quantitative PCR.The results showed that all the 91 samples of bioaerosol were found to have animal, plant and bacterial components, except that 3 samples were undetected plant-derived components, the positive rate was 96.7%.It demonstrates the feasibility of this method for the detection of bioaerosol samples and provides an effective means for environmental bioaerosol monitoring.Through the analysis of aerosol samples from different environmental air function areas in Beijing,Hangzhou and Wuhan,it is found that although most of the class II ambient air functional areas are not rich in vegetation cover,plant-derived components can still be found in their aerosol samples. And the content of plant components in the class II functional area in Hangzhou is higher than that in the class I functional area. The content of bacterial-derived components is significantly higher than that of animal and plant components, and the bacterial content in air samples in Beijing is higher than that in Hangzhou and Wuhan. The effects of environmental factors such as temperature, humidity and altitude on the bioaerosol content of the three cities were analyzed. It was found that the bacterial content in the air was negatively correlated with temperature, positively correlated with humidity, and positively correlated with altitude.Animal content is positively correlated with temperature and negatively correlated with humidityThis study creatively combines the housekeeping gene with bioaerosol detection to establish a method for detecting the content of bioaerosol components, and proved that the method has good stability and sensitivity through experiments. This study provides a new idea for the study of bioaerosols, and it is believed that it can lay the foundation for the establishment of environmental bioaerosol detection standards.
