| Chemotherapy is one of the commonly used methods in clinical cancer treatment.As an early and systemic treatment of cancer,chemotherapy has its unique advantages and irreplaceable status compared with surgical treatment,radiotherapy and immunotherapy.The development and testing of chemotherapy drugs are of great significance for cancer chemotherapy.Cell in vitro testing of chemotherapy drugs is an effective method for the development and screening of chemotherapy drugs.The electrical characteristics of cells are closely related to the physiological state of cells.The cell impedance measurement technology realizes the real-time nondestructive monitoring of the electrical characteristics of cells.Therefore,the effect of chemotherapeutic drugs on the state and function of tumor cells was studied by cell impedance testing.The second chapter,through the establishment of cell impedance test based on RC model equivalent circuit model of the parts of the cell impedance test to analyze the influence of the overall impedance,the results show that the system impedance characteristics change is affected by the coupling cell internal electrical parameters,based on the cell impedance sensor test cell function representation provides a theoretical basis.In chapter 3,we assembled a cell impedance sensor to analyze the effects of three chemotherapeutic drugs of different concentrations on the growth status of tumor cells.The results showed that the high concentration drug had a stronger inhibitory effect on cell growth than the low concentration drug,and cisplatin and doxorubicin could act on tumor cells faster than 5-fluorouracil.At the same time,by comparing the impedance data with the absorbance of CCK-8,it was found that the chemotherapy drugs may change the morphology and adhesion of tumor cells.In chapter 4,we further studied the effect of chemotherapy drugs on the adhesion of tumor cells by immunofluorescence staining of the adhesive plaque protein Vinculin,which is involved in the process of cell adhesion.The results showed that although the adhesion ability of tumor cells treated with chemotherapy drugs to re-adhere to the matrigel was reduced,the expression of tumor cell adhesion plaque protein Vinculin was up-regulated by chemotherapy drugs.Based on impedance testing technology,this paper provides a new method for in vitro testing of chemotherapy drugs,improves the effect of chemotherapy drugs on the function of tumor cells,and provides effective guidance for the development and screening of chemotherapy drugs,as well as the control of dose. |
