Crude rice bran sterol is the byproduct during the processing of rice bran oil,which contains sterol and triterpene alcohol.Sterol and triterpene alcohol could be widely used in medical,health care,food processing and some other fields.However,the crude rice bran sterol was almost discarded at present due to the limitation of the separation and purification technique.So we studied the separation of sterol and triterpene alcohol from crude rice bran sterol and the application of sterol and triterpene alcohol in this work.The brief research contents of the paper were listed as follows:1.The composition of crude rice bran sterol was determined by gas chromatography-mass spectrometry.We found that the content of sterol was 23.61%,the content of triterpene alcohol was 64.10%,and the content of other substances was 12.29%.2.We used thin-layer chromatography(TLC)to obtain sterol and triterpene alcohol from crude rice bran sterol.Then the yield and purity of sterol and triterpene alcohol influenced by the quantity of crude rice bran sterol and recycle number of silica gel column were discussed.With the increase of sample loading,we found that the yields of sterol and triterpene alcohol continued to be enhanced,and the purity continued to be decreased.The purity of the sterol and triterpene alcohol obtained from crude rice bran sterol by the TLC could be as high as 98%with a 13 cm effective length and an 8 cm effective diameter silica gel column under a 5 g input mass,and the yield of the triterpene alcohol and the sterol were 50%and 20%respectively.The effect of separation did not change significantly after the silica gel column reused three times.3.We changed the separation method as crystallization to obtain sterol and triterpene alcohol from crude rice bran sterol.Frist,we dissolved crude rice bran sterol as a solution,then we cooled the solution and we found that the sterol was enriched in the crystalline solid and the triterpene alcohol was enriched in the mother liquor.Ethanol was selected as the solvent for this project considered the requirements of industrial.We compared the differences between 95%(v%)ethanol(5%water content)and pure ethanol(water content less than 0.3%)as solvent respectively in the crystallization experiments.Combining all of the factors,95%(v%)ethanol was selected as the solvent.The effects of crystallization temperature and crystallization time and concentration on the yield and purity of the product were discussed using single-factor experiments and orthogonal experiments.We could obtain products with triterpene alcohol content of more than 83%in liquid and sterol about 35%in solid at-5℃ crystallization temperature with 8 hours crystallization time in 0.06 g/ml crystallization concentration.4.We explored the applications of sterols and triterpene alcohols in oil antioxidants.After adding 0%,0.05%,0.1%,0.15%,0.2%(wt.%)of sterol or triterpene alcohol to soybean oil,corn oil,peanut oil,and sunflower oil,we analyzed the antioxidant performance of these sterol or triterpene alcohol.We found that the addition of sterol or triterpene alcohol could enhance the oxidation resistance of oils.The antioxidant properties of oils were improved with the increase of sterol or triterpene alcohol added.We separated and purified the sterol and triterpene alcohol as 98%purity sample from crude rice bran sterol successfully with TLC and crystallization method,which were green and economic technology in industry.The sterol and triterpene alcohol could be used as statin,antiviral,antibacterial,antioxidants,and antitumors.We explored the application of sterol and triterpene alcohol in oil antioxidant,and found that sterol and triterpene alcohol could be used as the sacrificial agent in oil antioxidant.These results could be considered as the guidance researchment for the potential application of crude rice bran sterol.
