| 3-Amino-1,4-dimethyl-5H-pyrido[4,3-b]indole(Trp-p-1),a heterocyclic amino carcinogen,is widely created in food processing and cooking.Currently,it has been proved to be discovered in air,flue gas,barbecue and drinking water.Although its carcinogen ability in human has not been totally proved,its carcinogen ability in apes has been proved.Nowadays,more and more Trp-p-1 inhibitors have been discovered,such as flavonoids derivatives,methyl-xanthine compounds and some carbohydrate compounds with low moleculr weight.But,the research about these chemicals were less enough to effectively prove their Trp-p-1 absorption ability.Moreover,the Trp-p-1 absorption ability of these materials in vivo needs to be further confirmed.In order to search for better and reliable Trp-p-1 absorption inhibitors,we firstly used the Isothermal Titration Calorimetry to predict the binding energy between dietary fibers and Trp-p-1.Results showed that CMC-Na,Xantahan gum and Carrageenan have obvious molecular interations with Trp-p-1.Then,MDCK-MDR1 cell monolayer assay and in vitro drug absoruption model were used to evaluate the ability of diretry fiber in inhibiting the in vitro cell permeation of Trp-p-1.Results proved that CMC-Na,Xantahan gum and Carrageenan are effective in inhibiting the in vitro cell permeation of Trp-p-1.Following this,pharmacokinetic experiments were used to evaluate the ability of dietary fibers in inhibiting the Trp-p-1 absorption in blood.CMC-Na,Xantahan gum and Carrageenan could significantly reduce the value of Cmax,AUC(O-t),AUC(0-∞)of Trp-p-1.To sum up,dietary fibers could be used as Trp-p-1 absorption inhibitors and possibly reduced its carcinogen ability... |
