| Objective: Hawk tea is a kind of plant substitute tea made from the tender buds and leaves of leopard skin camphor,which are processed by greening,rolling,and frying.Antioxidant,antibacterial,lowering cholesterol,protecting liver,improving immunity and other biological activities.Our country’s hawk tea has a wide geographical distribution and rich resources.It is often sold in the form of agricultural and sideline products,and its deep-processed products are relatively few.As a kind of medicine and food homologous material,hawk tea is worthy of in-depth research and development for its medicinal value and health care value.In this paper,hawk tea was used as raw material,flavonoids were extracted from it,selenomethionine,methylselenocysteine,sodium selenite and hawk tea flavonoids were used as research objects to compare the effects of single component and compound component on camellia tea.The synergistic antioxidant effect of oil,Idesia polycarpa Maxim.oil and lard;and taking liver cancer cells as samples to explore their effects on the proliferation activity of liver cancer cells.By researching and developing the application technology of selenium in oil,developing new substances of natural antioxidants in oil,realizing scientific supplementation of selenium,delaying the oxidation of oil and improving its use value and laying a theoretical and technical foundation.Methods: The total flavonoids of hawk tea were extracted with ethanol,purified by static adsorption on macroporous resin D101,and then scanned by infrared spectrum to identify the structure of the extract.The change of peroxide value of oil within 16 days was determined by schaal oven method and sodium thiosulfate titration method.,to determine the antioxidant effect of antioxidants;in Hcclm3 cells,the synergistic effects of selenium and total flavonoids of hawk tea on cell proliferation rate and migration rate were determined by CCK-8 method and cell scratch method.Results:（1）The characteristic functional groups in the infrared spectrum of the ethanol extract of hawk tea were analyzed as flavonoids.Taking IVIdesia polycarpa Maxim.oil as the research object,by comparing the peroxide value of oil,the best choice of emulsifier is 1.2% monoglyceride,and the optimal amount of total flavonoids in hawk tea is 0.015g/30 g.（2）When selenomethionine,methylselenocysteine,sodium selenite and total flavonoids of hawk tea act on camellia oil separately,they have obvious antioxidant effects.the stronger.The antioxidant effects of selenomethionine,methylselenocysteine,and sodium selenite on camellia oil after compounding with the total flavonoids of hawk tea were better than those of single-component additions,indicating that the compounding of selenium and flavonoids had an effect on camellia tea.Oil antioxidants have a certain synergistic effect.When selenomethionine,methylselenocysteine,sodium selenite and total flavonoids of hawk tea acted on lard separately,the antioxidant effect was obvious.When selenomethionine and flavonoids were compounded,the POV values??of lard from high to low were in the order of selenomethionine and total flavonoids in hawk tea compound group>flavonoid control group>blank group>selenomethionine alone group>TBHQ group;When sodium selenate and flavonoids were compounded,the POV values ??of lard from high to low were in the order of sodium selenite and hawk tea total flavonoids compound group>flavonoid control group>blank group>sodium selenite alone group>TBHQ group;When methylselenocysteine ??and flavonoids were compounded,the POV value of lard from high to low was in the order of methylselenocysteine ??and hawk tea total flavonoids compounding group>flavonoid control group>blank group>Methylselenocysteine ??alone group>TBHQ group.The results showed that within the concentration range of this experiment,selenium and total flavonoids of hawk tea did not have a synergistic effect on lard,and even promoted the oxidation of oil.When selenomethionine,methylselenocysteine,sodium selenite and total flavonoids of hawk tea acted on kaempferia oil alone,they had obvious antioxidant effects.When selenomethionine and flavonoids were used in combination,the POV values??of Idesia polycarpa Maxim.oil from high to low were blank group > flavonoid control group > selenomethionine and hawk tea total flavonoid compound group >selenomethionine alone group > TBHQ group;When methylselenocysteine ??and flavonoids were used in combination,the POV value of Idesia polycarpa Maxim.oil from high to low was blank group > methylselenocysteine ??combined with total flavonoids of hawk tea > single addition Methylselenocysteine ??group>flavonoid control group>TBHQ group;when sodium selenite and flavonoids were used in combination,the POV value of kaempferia oil from high to low was blank group>sodium selenite and hawk tea Total flavonoid compound group>sodium selenite alone group>flavonoid control group>TBHQ group.Comparing the antioxidant effect of Idesia polycarpa Maxim.oil,the POV value of the compound group was higher than that of the single component,indicating that within the concentration range of this experiment,selenium and hawk tea total flavonoids did not have a synergistic effect.Taking liver cancer cells as the research object,after the three selenium compounds acted synergistically with the total flavonoids of hawk tea,the proliferation inhibition rate of Hcclm3 cells could reach more than 90%,showing extremely high tumor suppressor activity.In particular,when methylselenocysteine??is used in combination with the total flavonoids of hawk tea,the inhibition rate can still reach 83% at a very low concentration such as 78.1 μg/m L.Selenium and total flavonoids of hawk tea have obvious inhibitory effect on the migration of Hcclm3 cells.The results showed that selenium and the total flavonoids of hawk tea synergistically inhibited the proliferation and migration of cells.Conclusion:Selenium and hawk tea total flavonoids synergistically act as antioxidants for oils and fats,and have a good synergistic effect on camellia oil,but have no obvious effect on lard and Idesia polycarpa Maxim.oil,and the compounding method needs to be improved.The synergistic effect of selenium and the total flavonoids of hawk tea on the proliferation and migration of liver cancer cells is remarkable. |
