Cancer has a serious impact on people's health and life.Once cancer is contracted,it will cause huge physical and economic burden to individuals and families.Therefore,it is necessary and urgent to research and develop effective drugs for cancer treatment.In addition,scientists have found that liver cancer and lung cancer often do not respond well to chemotherapy and are prone to drug resistance,so the search for safer and more effective treatments must continue.Nanotechnology is rapidly developing in the field of public health and has become one of the most powerful tools for disease prevention,diagnosis and treatment.Boron nitride(BN)nanomaterials have the advantages of good biodegradation,antibacterial,anti-tumor inhibition and so on.Because of these advantages,BN has gradually become a hotspot in the field of biomedical research in recent years.Due to the unique edge structure of two-dimensional BN nanosheets,it has potential application value in the biomedical field.In this work,three sizes of BN nanosheets were selected to study the effect of size on tumor inhibition and safety.By TEM,SEM and particle size analysis,the sizes of BN-a,BN-b and BN-c nanosheets were determined to be about 130 nm,770 nm and 3.2?m,respectively,and these three kinds of BN nanosheets were effective against human hepatoma Hep G2 cells The inhibitory effect of human hepatoma cell Huh7 and human lung cancer cell A549 was studied and compared with human normal liver HL-7702.It is found that BN nanosheets have typical h-BN characteristic structure,and the content of B sustained release gradually increases with the increase of time and temperature,but it basically reaches the saturation state in 1-2days.BN-b and BN-c nanosheets reduced the survival rate of Hep G2 cells by 36%and 30%,and BN-a,BN-b and BN-c nanosheets reduced the survival rate of Huh7 cells by 69%,46%and 29%,BN-a nanosheets showed slight cytotoxicity to human normal hepatocyte HL-7702cells,and the cell survival rate of BN-a,BN-b and BN-c nanosheets to A549 cells decreased by 52%,53%and 43%.Hep G2 and Huh7 cells were found to be sensitive to BN nanosheets size,HL-7702 and A549 cells were found to be insensitive to BN nanosheets size,and Hep G2,Huh7 and A549 cells were found to be dose-dependent.The morphological changes of Hep G2,Huh7,HL-7702,A549 cells induced by BN nanosheets were independent of size.The cell morphology of Hep G2 cells was not significantly affected,but Huh7,HL-7702 and A549 cells were affected.The effect of BN nanosheet on apoptosis of Hep G2,HL-7702,A549 cells was relatively small,and that of Huh7 cells was relatively high.On the basis of the above research,the magnetic boron nitride composite(Fe3O4/BN-a)with a particle size of about 6.6 nm was prepared by mixing BN-a nanosheets with ferric acetyl acetonate through in-situ reduction method,using the good biocompatibility and magnetic properties of iron oxide to obtain functional boron nitride and study its tumor inhibition and safety evaluation.Fe3O4/BN-a composite showed a dose-dependent inhibitory effect on Hep G2 Huh7 and A549 cells,and had no toxicity to HL-7702 cells.Compared with BN-a nanosheets,Fe3O4/BN-a composite showed enhanced inhibitory effect on Hep G2 and A549cells,weakened inhibitory effect on Huh7 cells,and enhanced inhibitory effect on A549 cells.The survival rate of Hep G2 cells decreased to about 58%,and the inhibitory effect on A549cells increased by about 2%.Hep G2 and HL-7702 cells were not significantly affected,but Huh7 and A549 cells were affected.The apoptosis rate of Hep G2,Huh7 and A549 cells was increased,but not HL-7702 cells.Magnetic BN nanosheets did not significantly change the inhibitory effect of BN nanosheets on cancer cells and normal cytotoxicity,indicating a certain feasibility,but the actual application still needs to be tested and studied according to cell types.In conclusion,BN nanosheets is relatively non-toxic to normal liver cells,and has a certain inhibitory effect on liver cancer cells with size and dose dependent effect.BN nanosheets has certain application value in tumor inhibition research,and it is worth further exploring the influencing mechanism.
