Objective:To fabricate the composite nanoparticles that synthesized dual-mode imaging and multifunctional therapy(hydrogen,photothermal,and ultrasound therapy)and evaluate their prospect as nanocarriers to integrate oncology diagnosis and treatment.Methods:We compounded Pd H2/Au@PFH@MPDA(PHAPM)nanoparticles,which contained Pd/Au,H2,and PFH as the core and MPDA as the stabilizing shell.The corresponding machine detected their morphology,diameter,the electric potentials.We also investigated the photothermal capacity,hydrogen release property,and the liquid-gas phase transition of PHAPM.The ultrasound and photoacoustic imaging ability of PHAPM were evaluated in vitro.Furthermore,the cytotoxicity of PHAPM and the killing effect of PHAPM combined with multifunctional therapy(hydrogen,photothermal,and ultrasound therapy)on MDA-MB-231breast cancer cells were evaluated in vitro.Results:The PHAPM nanoparticles were spherical with the diameter of 110.3±8.5nm,and the average potentials was-16.7±0.4m V.MDA-MB-231 breast cancer cells were combined with PHAPM for 4 hours to achieve optimal uptake.The temperature of 150?g/m L PHAPM could rise to 66.5±0.4?after 808nm NIR irradiation for 10 min.Methylene blue and ultraviolet spectrum experiments confirmed that PHAPM successfully loaded hydrogen and could accelerate the release of hydrogen under NIR irradiation.With the increase of temperature,PFH underwent liquid-gas phase transition,the ultrasound echo of PHAPM gradually increased.The photoacoustic value gradually increased with the higher the PHAPM concentration.The PHAPM had good biocompatibility with no obvious cytotoxicity.And the PHAPM treated with NIR and ultrasonic irradiation showed the strongest cell-killing effect.Conclusion:The PHAPM nanoparticles have dual-mode imaging and have multi-mode tumor therapy functions,which are expected to provide a new method for the integration of tumor diagnosis and treatment.
