As a large agricultural country,China has a vast territory and abundant products.At the same time,China is also a large population.In order to solve the food problem and ensure the yield and quality of crops,the use of pesticides is essential.Up to now,there are more than thousands of pesticides and veterinary drugs,which makes it more and more difficult to detect food safety,while drug abuse,illegal addition and other behaviors make it more difficult to screen harmful substances in food.Therefore,in this study,pesticides,veterinary drugs and their metabolites were selected as the research objects,aiming at the outstanding problems such as the small capacity of the existing liquid chromatography-high resolution mass spectrometry library and the lack of information covering small molecular compounds,resulting in large uncertainties in non-directed screening,low identification accuracy and poor applicability.Build food chemical hazard factor and its metabolic product standard chromatography with high resolution mass spectrometry screening database,and it is difficult to obtain standard material of agriculture veterinary drug metabolites in liver microsomal incubation in vitro biotransformation experiments,using mass spectrometry analysis of metabolites,increasing the database is also provide data basis for study of the drug toxicity.The main research results are as follows:1.Qexactive Plus technology was used to establish a standard mass spectrometry database of agricultural and veterinary drug hazard factors in food,including mass spectrometry database and information text database.The database contained 1084 chemical hazard factors,including 757 pesticides and 327 veterinary drugs,and a total of1084 first-level mass spectrograms and 3252 second-level mass spectrograms were obtained.In order to improve the ability of non-directed screening of food chemical hazard factors in China.2.The injection and addition modes of standard substances were summarized.For pesticides and veterinary drugs,the hydrogenation peak was mainly detected by mass spectrometry,supplemented by ammonia peak and sodium peak and hydrogen reduction peak;salt and hydrate standard substances would be hydrogenated or dehydrogenated after desalting and dehydrating.The fragmentation situation under different collision voltage was analyzed by taking the thiocarbate pesticide artemisinil and the fluoroquinolone antibiotic enfloxacin as examples,and the possible fragmentation path map of the two substances was obtained,which helped remove background interference peak,enhanced screening ability,and provided data for the qualitative and quantitative detection of related drugs.3.With new neonicotinoid pesticide imidacloprid,usage in organism worm amidine as a substrate for liver microsomal temperature hatch in vitro studies,using mass spectrometry identification of metabolites,received a total of six metabolites,including5 kinds of imidacloprid metabolites,1 kind of organism worm amidine metabolites,and speculated that the formula for the concrete analysis of metabolites of metabolic pathways,The metabolite information was added into the database,which also provided the idea data for the study of neonicotinoid pesticide toxicology.
