Grape planting areas of China and produce remain the first places all over the world.However,a large number of by-products will be produced in the processing of grapes and grape seeds are in a relatively large proportion in the by-products.But grape seeds can't be fully used which restricts development of grape seeds industry because of various reasons.(1)Stepwise extraction of protein and soluble dietary fiber from grape seed meal.Five enzymes were selected,and papain was selected under the optimum enzymatic hydrolysis conditions with the extraction rate as the index.Cellulase was used to extract soluble dietary fiber,and single factor and response surface experiments were used to extract soluble dietary fiber.The optimum technological parameters are determined as follows:The protein extraction rate was 80.22%with the addition of 1.5%,pH 7.0,temperature 50? and time 40 min.The extraction rate of soluble dietary fiber was 58.29%with the addition of 2.00%,pH 5.0,enzymatic hydrolysis temperature 55? and enzymatic hydrolysis time 60 min.(2)Synchronized extraction of protein and soluble dietary fiber from grape seed meal by complex enzymatic method.Papain and cellulase were compounded in a ratio of 3:1,using single factor and response surface methodology.The optimum technological parameters are determined as follows:The extraction rate of protein and soluble dietary fiber from grape seed meal was 78.36%and 57.21%respectively under the conditions of pH 6.0,enzymatic hydrolysis temperature 50? and enzymatic hydrolysis time 50 min.(3)The spray drying technology of grape seed meal extract was optimized by single factor test and orthogonal experiment.Determine the primary and secondary relationship of each factor:Outlet temperature>atomizer speed>inlet temperature.The system air temperature is 160?,the outlet air temperature is 75? and the atomizer speed is 55 Hz.Under these conditions,the best spray drying effect is achieved.(4)The digestibility and absorptivity of grape seed meal extract were determined by digestion experiment in vitro.The results showed that the digestibility and absorptivity of grape seed meal protein were higher in gastric environment.In intestinal neutralization and total digestive absorption,the absorptivity of grape seed meal extract was significantly higher than that of grape seed meal protein(p<0.05).The results showed that the swimming time of mice in three different dose groups was significantly longer than that in the blank group(p<0.05),the content of hepatic glycogen was increased,and the content of serum urea nitrogen and blood lactic acid was significantly decreased(p<0.05).Compared with the model group,the propulsive length,propulsive rate,the time of first black stool defecation and the weight of feces in the three dose groups were significantly different(p<0.05).
