Guava is a nutritious tropical fruit,rich in natural active ingredients such as polyphenols,polysaccharides,and peptides,and has high nutritional value.T his thesis takes the red-heart guava pomace after extracting free polyphenols asthe research object,applies the response surface test design,uses the high-volt age pulsed electric field method to optimize the extraction conditions of the co mbined polyphenols;in the animal experiments,the metagenomic sequencing te chnology is used to detect The diversity,richness,composition and relative abu ndance of the intestinal flora of mice after ingesting guava containing bound p olyphenols,and found that bound polyphenols have a regulatory effect on the i ntestinal flora of mice.The main research results of this paper are as follows:(1)The high-press ure pulse electric field is used to extract the combined polyphenols,and the o ptimal extraction conditions are: 200 pulses,field strength 8.2 kv/cm,material-l iquid ratio 3:1 mg/m L,and the measured combination The actual yield of poly phenols is 6.416 mg/g.(2)Compared with the blank group,after the intragastric administration of polyphenols,the diversity and richness of the bacterial group in the low conce ntration group increased(P<0.05),and the diversity and richness of the bacteri al group in the high concentration group showed a downward trend(P<0.05),which proves that low concentration of combined polyphenols can promote the diversity and richness of the flora,while high concentration can inhibit the div ersity and richness of the flora.(3)By sequencing all microorganisms in the intestinal contents of mice,a total of 10 phylum levels were detected.Mainly includes Firmicutes,Bacteroide tes,Verrucomicrobia,Proteobacteria,Actinobacteria,Deferribacteres,Epsilonbacte raeota,Cyanobacteria,Patescibacteria,Tenericutes.Among them,the relative ab undance value of the three phylums of Firmicutes,Bacteroides and Verrucomicr obia is the largest,and the sum of the three is more than 95%.The high con centration of bound polyphenols will reduce the number of pathogenic bacteria,while the low concentration of bound polyphenols has a greater impact on the ratio of the thick-walled phylum and the Bacteroidetes.(4)Mouse feces identified 96 genera from 10 phylum levels.Genera with relative abundance greater than 0.1% accounted for 98.79% of the total commu nity.Among them are the three genera of Akkermansia belonging to Verrucomi crobia,Lactobacillus of Firmicutes,and Bacteroides of Bacteroidetes.Has the h ighest relative abundance.During the intragastric administration of polyphenols,the blank group,the relative abundance growth rate of Akkermansia was 35.921%;the low-dose group,the relative abundance growth rate of Akkermansia w as 102.012%;the high-dose group,the relative abundance growth rate of Akker mansia was 387.082%.Compared with the blank group,the low-dose combinati on of polyphenol treatment reduced the relative abundance of Akkermansia,an d the high-dose combination of polyphenol treatment increased the relative abu ndance of Akkermansia.The differences were not significant,which may be In dividual factors in mice have led to an increase in the basic differences in inte stinal flora and affected the consistency of the results of the study,but it is n ot difficult to find through comprehensive analysis that the higher the concentr ation of guava pomace gavage preparation,the faster the relative abundance of Akkermansia increases.
