Placenta is a kind of high-quality resource with rich natural ingredients,good nutrition and various biological activities.New Hope Group,Every year sows give birth to about 300 thousand babies in 2018.If 3 kg placenta is collected per sow,900 tons placenta can be used for processing every year.Therefore,the supply of pig placenta is not only sufficient,but also low cost.In fact,pig placenta is often abandoned,resulting in waste of resources.The purpose of this paper is to study the antioxidation and immunity enhancement of pig placenta,optimize the freeze-drying process of pig placenta,and enhance the added value for the processing of pig placenta in the future.The specific research contents are as follows:(1)Component analysis,antioxidation and immunity enhancement of fresh pig placenta:the basic components of fresh pig placenta were determined,the antioxidation ability of fresh pig placenta extract was evaluated by cytology,and the immune ability of pig placenta extract was determined by injecting pig placenta extract into mice.The results of basic component analysis showed that the content of protein in fresh pig placenta was about 8.28%,the content of calcium and phosphorus were 3.05 and 537.52 mg/kg,respectively.,and the moisture content is more than 90%.The results of antioxidant capacity showed that 0.8?g/mL of pig placenta extract could reduce the mortality of LO2and HaCaT cells,the cell viability of LO2 cells in 48 h was 11.1%higher than that of the control group,and the cell viability of HaCaT cells in 24 h was 11.2%higher than that of the control group,with significant difference between the two groups Compared with the control group.LO2 and HaCaT cells treated with 0.8?g/mL pig placenta extract had significant difference in their anti-H2O2oxidative damage ability.The mice were injected with 0.4 mL extract for 7 d showed that the immune organ index,macrophage phagocytosis rate and T-lymphocyte transformation rate of male mice increased significantly,while that of female mice was only significantly different in T-lymphocyte transformation rate.(2)Preparation and functional activity analysis of freeze-dried powder of pig placenta:taking appearance and antioxidant capacity as inspection indexes,through single factor experiment,the optimal concentration of freeze-dried protective agent is 4 mg/mL(sucrose),freeze-drying time is 24 h,and freeze-drying thickness is 0.4 cm.Taking the antioxidant capacity as the index,the orthogonal experimental design was carried out for three factors:the concentration of protectant,the time of freeze-drying and the thickness of freeze-drying The factors affecting the cell viability were determined as follows:sucrose concentration>freeze-drying time>freeze-drying thickness.The best technological conditions were 6%of protectant concentration,24 h of freeze-drying time and 0.6 cm of freeze-drying thickness.By functional activity analysis,the antioxidant capacity and immune function of freeze-dried pig placenta were preserved,and the content of amino acid and protein was about 10 times that of fresh pig placenta.(3)The compatibility optimization and toxicology research of pig placenta powder capsule preparation:Taking pig placenta freeze-dried powder as the main raw material,supplemented with bee pollen,licorice,American ginseng and other precious Chinese medicine,using 5 factors and 4 levels of orthogonal experimental design.16 pig placenta powder preparations were obtained.Kunming mice were taken as the experimental object,and the pharmacology experiment was carried out.The results showed that:pig placenta powder,American ginseng,Chinese medicine C,Chinese medicine D and bee pollen is200:25:4.5:10:80 of the best ratio o,which has obvious effect on immune organs and immune cells of mice.Meanwhile,this ratio can delay the aging of mice.The results showed that the LD50of pig placenta powder was more than 19200 mg/kg.The results showed that there was no effect of pig placenta powder on sperm distortion.
