Wheat(Triticum aestivum L.,AABBDD,2n=42)is one of the three important food crops in the world.It has been cultivated for more than 8,000 years and plays an important role in the production and development of world agriculture.Wheat in human food and clothing as well as to provide the nutrition for human,people’s attention on wheat food quality also with the development and progress of science and society,increasingly intense,the discovery of wheat gliadin sensitization and challenge to strengthen the creatures of the wheat in the world,health grain project work such as promoting the nutrition and health has become a nearly 10 years international wheat quality research topic.In the broad sense,wheat quality includes processing quality and nutritional quality,and some fields also include morphological quality.Nutritional quality includes balanced content and proportion of proteins,carbohydrates,lipids and some enzymes.In recent years,arabinoxylan in wheat has attracted more and more attention due to its biological activities of dietary fiber,such as lowering cholesterol level,regulating blood sugar,antioxidant and possible anti-tumor and immunomodulatory effects.The effect of polyphenol oxidase on the color of wheat flour is directly related to its commercial characteristics and nutritional value.Three recombinant inbred line populations were used in this study,which were(1):R146×R316;(2): yanzhan 1(YZ1)×J1809;(3): yanzhan 1(YZ1)× R97 / R146,planted in the qionglai wheat breeding base of sichuan agricultural university in 2017-2018.The contents of each protein component,arabinoxylan and polyphenol oxidase activity were determined.The alcohol-soluble proteins of R146×R316 were analyzed by a-page electrophoresis,and the results were as follows:1.Determination of protein content of each material: proteins of each component of three sets of materials were determined,and the results showed that there were significant differences among these materials.(1)the two-year data of material no.1 were relatively stable,and the average contents of globulin,alcohol-soluble protein and total protein were not significantly different,indicating that the accumulation of globulin and alcohol-soluble protein of the material was less affected by environmental factors.2.used the A-PAGE electrophoresis method to analyze the seed gliadin strip of(1)R146×R316 material.The results showed that in material(1),a total of 30 bands were isolated by gliadin,with the lowest total band number of 14 bands and the highest 28 bands.The correlation analysis of the number and content of bands in each protein region showed that the content of alcohol-soluble protein was positively correlated with the number of bands isolated from each protein region.By 2018,the content of alcohol-soluble protein in this material was significantly correlated with the number of bands in the β protein region,and significantly positive correlation with the total number of bands.For 2019,the content of alcohol-soluble protein in this material was positively correlated with the number of bands in the α protein region,and significantly positively correlated with the total band number.For two years,the average content of alcohol-soluble protein was positively correlated with the number of bands in the α、β protein region and positively correlated with the total band number.3.Determination of Arabian xylan content: the determination of Arabian xylan content in three sets of materials showed that there were significant differences among these materials,and the content of material no.(1)was significantly lower than that of material no.(2)and(3).(1)the data of material no.1 was stable in two years with no significant difference,but the number of offspring with higher content increased compared with that of the previous year.4.Determination of polyphenol oxidase activity: the results of the determination of polyphenol oxidase activity in three sets of materials showed that there were significant differences among these materials,and the content of material no.(1)was significantly lower than that of material no.(2)and(3).(1)the data of material no.1 is stable in two years and the difference is not significant.5.analysis of the above three angles and previous experimental results shows that from the point of view of reducing the content of gliadin to reduce the risk of sensitization,there are the lowest content offspring in the YZ1/J1809 of recombinant inbred line population,which has the potential to screen low content gliadin materials.From the point of view of increasing the content of arabinoxylan to enhance the health function of wheat,the recombinant inbred population YZ1/J1809 、 YZ1/R97/R146 suitable for screening experimental materials with high content of arabinoxan.from the point of view of commodity characteristics,R316/R146 recombinant inbred population showed the best performance,and the offspring with enzyme activity below 40 AU accounted for the majority.6.The materials with very low alcohol-soluble protein content(& lt;2.7%),high arabinoglycan content(more than 8%)and low polyphenol oxidase(less than 40 AU)were finally screened in the high-generation recombinant inbred line material R316/R146.Five progeny were screened in this set of materials,numbered 154,161,166,167,224.The electrophoresis bands of these materials are similar,the missing bands in ω region are more,and the missing bands are concentrated in the ω region close to the γ region.At present,the research on grain polyphenol oxidase activity of common wheat in China is still lagging behind that of developed countries,and the research progress on polyphenol oxidase activity and wheat quality improvement is slow.Therefore,for wheat with different genetic background,the distribution and genetic characteristics of polyphenol oxidase activity are studied.
