| Cancer has become one of the major chronic diseases threatening human health.Traditional treatment methods,such as surgery,radiotherapy,and chemotherapy,still have disadvantages such as large side effects,low treatment efficiency,and easy recurrence.In recent years,in order to overcome the shortcomings of traditional treatment methods,some emerging cancer treatments based on nanomaterials have appeared.Nano-metal organic frameworks(NMOFs)have received more attention in the biomedical field because of their large specific surface area,good biocompatibility,adjustable pore structure and diverse functions.Derivatives based on NMOFs can not only be used as imaging agents and drug carriers,but also can use their enzyme-like activities to assist cancer treatment.Based on this background,this paper synthesizes metal-organic framework materials and their derivatives,exploring their ability to load,effectively release,and enzyme-like activities,so as to provide theoretical guidance for new tumor treatments.The specific research content is as follows:(1)First,we prepared nano-metal organic framework-MIL101(Fe)nanoparticles through a simple hydrothermal synthesis method.Based on their large surface area and pore structure,they can be loaded with guest molecules 3,3ˊ,5,5ˊ-tetramethyl Benzidine(TMB).The Fe ions released by MIL101(Fe)in the micro acid environment(TME)of the tumor catalyze the generation of highly toxic hydroxyl radicals(·OH)from the endogenous hydrogen peroxide(H2O2)of tumor through the Fenton effect,thereby inducing cells Apoptosis;at the same time,·OH further oxidizes the loaded TMB.The oxidation state ox TMB can absorb near-infrared light and convert it into heat,causing the tumor to heat up locally,thereby causing tumor cells to ablate.The heat generated further enhances the production of·OH.Therefore,the nano-construct based on MIL101(Fe)-TMB utilizes the synergy of chemical kinetics and photothermal(CDT/PTT)to greatly improve the efficiency of cancer treatment.(2)Secondly,we quickly obtained bimetal oxide(CoFeOx)by laser marking metal organic frame material(MOF-74-Co/Fe).We use hyaluronic acid(HA)to modify its surface(CoFeOx-HA)to improve hydrophilicity.CoFeOx-HA has the catalytic properties of oxidase(OXD enzyme),superoxide dismutase(SOD enzyme)and peroxidase(POD enzyme)under TME.Oxygen can be converted into superoxide anion(·O2-)by OXD enzyme.·O2-disproportionates to generate H2O2 under the catalysis of SOD enzyme,and the generated H2O2 is further catalyzed by POD enzyme to highly toxic·OH.Under the condition of low concentration of H2O2,there is still the ability to produce ROS,thus realizing anti-cancer treatment. |
