"Jinghe wolfberry" is one of the first batch of geographical indication products mutually recognized by China and the EU.Due to its unique natural ecological environment advantages,"Jinghe wolfberry" has formed a unique nutritional quality.In order to fully identify the nutritional quality characteristics of "Jinghe wolfberry" and clarify the characteristic fingerprints of amino acids,mineral elements,sugars and organic acids in Xinjiang "Jinghe wolfberry" and Ningxia "Zhongning wolfberry",this paper uses Xinjiang "Jinghe wolfberry" With Ningxia "Zhongning wolfberry" as the research object,the automatic amino acid analyzer was used to detect the content of 17 amino acids in wolfberry;the inductively coupled plasma mass spectrometer(ICP-MS)was used to detect the content of 52 elements in wolfberry;the use of high performance liquid chromatography Methods such as determining the content of main sugars and organic acids;combined with analysis of variance,discriminant analysis and correlation analysis,the systematic study of the characteristic fingerprints of nutritional quality differences in wolfberry from different origins;the study has drawn the following main conclusions:(1)The content of Ala and Pro in "Jinghe wolfberry" in Xinjiang is higher than that in "Zhongning wolfberry" in Ningxia,and the rest Asp,Thr,Ser,Glu,Gly,Cys,Val,Met,Ile,Leu,Tyr,Phe,His,Lys,Arg content of 15 amino acids is lower than Ningxia "Zhongning wolfberry".Among the 17 amino acids of wolfberry,the contents of the 5 amino acids,Asp,Val,Tyr,Phe,and Lys,have significant regional differences between the two producing areas;through VIP value optimization,the 17 amino acids are reduced to Cys,Lys,Tyr,His,Phe,Val,Gly,Ile 8 kinds of amino acids.(2)There are significant differences in 26 mineral elements between "Jinghe wolfberry" and "Zhongning wolfberry".Among them,"Jinghe wolfberry" Mg,Al,P,K,V,Cu,Zn,As,Se,Mo The content of 16 elements including,Ag,Cd,Gd,Er,Pb and U are significantly higher than "Zhongning wolfberry";OPLS-DA(orthogonal partial least squares method)established Xinjiang "Jinghe wolfberry" and Ningxia "Zhongning wolfberry" ”Discriminant model,optimize the number of elements in the wolfberry sample through the VIP value of the model,and reduce the model variables from 46 elements to 14 elements(U,Mo,K,Rb,Au,Ir,Hf,Cu,Tl,Zn,Ca,Th,Mg,Cd).(3)The main carbohydrates contained in wolfberry fruit are glucose,fructose and polysaccharides,and the order of their content in descending order is glucose>fructose>polysaccharides.The glucose and fructose content of Xinjiang "Jinghe wolfberry" samples was higher than that of Ningxia "Zhongning wolfberry" samples,and the polysaccharide content was lower than that of Ningxia "Zhongning wolfberry" samples.The results of variance analysis of sugar utilization of wolfberry from different producing areas showed significant differences in the content of polysaccharides;using OPLS-DA to screen out polysaccharides is a significant variable with VIP>1.(4)The main organic acids contained in the fruits of "Jinghe wolfberry" in Xinjiang are: citric acid,tartaric acid,malic acid and succinic acid,the content of which is citric acid>tartaric acid>malic acid>succinic acid.The content of citric acid and succinic acid in the samples of "Jinghe wolfberry" in Xinjiang is higher,and the contents of total acid,tartaric acid and malic acid in the samples of "Zhongning wolfberry" in Ningxia" are higher.The contents of malic acid and succinic acid are extremely significant differences between the two producing areas;the use of OPLS-DA to screen out malic acid and total acid from different producing areas of lyciic acid is a significant variable with VIP>1.(5)Finally screened out 8 kinds of amino acids(Cys,Lys,Tyr,His,Phe,Val,Gly,Il),14 kinds of elements(U,Mo,K,Rb,Au,Ir,Hf,Cu,Tl,Zn),Ca,Th,Mg,Cd),polysaccharides,malic acid and total acid are significant variables.In summary,no matter from the composition of amino acids and mineral elements,or from the composition of main sugars and organic acids,Xinjiang "Jinghe wolfberry" has its unique nutritional quality characteristics,such as Ala,Pro,fructose,glucose,The content of 16 mineral elements such as citric acid,succinic acid,K,Mg,Zn are significantly higher than that of "Zhongning wolfberry",and the accuracy of the identification of origin based on multiple indicators combined with stoichiometric methods can reach more than 90%.
