In the world,the demand for deer products in New Zealand,Australia and European countries is mainly venison,and the by-products are exported to Asian countries.The demand for deer products in Asian countries such as China,South Korea and Japan is deer by-products,such as antler,deer blood,deer fetus,deer bone and deer whip.In 2010,the Chinese Pharmacopoeia stipulates that sika deer and red deer products are traditional Chinese herbal medicines in my country,and are known as the top animal medicines.The main production areas are in the three northeastern provinces,and the antler with the largest demand for products is called the "Three Treasures of Northeast China."Due to changes in people's consumption habits,deer products are sold in the market in the form of processed products such as slices,granules,powder,and capsules.Because of the varying degrees of changes in product morphology and physical and chemical properties,it is difficult for consumers to judge the quality of products by conventional methods.In addition,the prices of different types of deer products(such as sika deer,red deer,red deer and reindeer)vary greatly in the market.Driven by profits,a large number of low-value products such as reindeer and red deer have flowed into my country,imitating the high-value Chinese medicinal materials sika deer and red deer products prescribed by the National Pharmacopoeia.In response to the above-mentioned market demand,this study selected the CEP295 L gene based on the evolutionary conservation characteristics of deer animals.CEP295 L exists as a single copy gene,and there are multiple conserved SNP sites among different deer species.This gene is used as a target to design three sets of primers for detection.The three sets of primer systems can identify the source of deer products.The main research results are as follows:The first set of CerF1R1 primers can distinguish whether the product is of deer origin.This system has a specific single band at Maker 540 bp for all kinds of different deer samples,and is not sensitive to other species such as cattle,sheep,and pigs..This primer system can quickly detect whether the product is a deer product.The second set of LuF2R2 primers can identify whether the deer products are high-value sika deer and red deer products.Under this system,sika deer and red deer samples have a specific single band at Maker 240 bp,and red deer samples have a specific double band between Maker100 bp and 240 bp.,Other deer breeds,cattle,sheep and pigs are not sensitive.This primer system can accurately identify whether deer products are derived from high-value sika deer or red deer,or low-value red deer or other deer products.The third group of SusFR and Ny FR primer identification systems are pig-specific primers and cow and sheep co-primers.Porcine primer(Sus FR)has a specific single band at the Maker480 bp position of all kinds of pig products,and there is no sensitivity to deer,bovid and ovine samples.The cattle and sheep co-primer(Ny FR)system has a specific single band at the Maker380 bp position for cattle and sheep products,and it is not sensitive to samples from the family Suidae and Deer family.This primer system can accurately detect whether deer products are adulterated with low-value cattle,sheep and pig products,and the experimental results are completely reproducible.This study established a set of molecular identification system for the detection of deer products by screening deer conserved genes,designing specific primers and exploring the best reaction conditions for PCR.This system can help people quickly,accurately and efficiently identify the provenance of deer products,provide a reliable scientific basis for product pricing,maintain the order of the deer product market and protect the rights and interests of deer farmers.This system can help people quickly,accurately and efficiently identify the provenance of deer products,provide a reliable scientific basis for product pricing,maintain the order of the deer product market and protect the rights and interests of deer farmers.
