Biological amines play an important role in the biomedical field and the food industry.They are considered to be important biomarkers for monitoring food quality and assisting diagnosis of diseases.In response to the explosive growth of food safety issues,especially in developing countries,simple,fast,low-cost and accurate food safety monitoring systems are becoming more and more urgent and indispensable.Click chemistry reaction has the advantages of higher yield,strong stereoselectivity,shorter reaction time,and reaction modularity.Its reaction conditions are simple,and theoretically all reactants can be converted into products,which conforms to the concept of atom economy and green synthesis.Besides the advantages of click reaction,the fluorescent probe based on click reaction also has many advantages,such as high design flexibility,easy to meet the detection requirements of people and so on.Therefore,it is necessary to design such a fluorescent probe based on click reaction to detect biogenic amines.On the basis of a large number of previous studies,three alkyne fluorescent compounds were synthesized based on three different fluorophores,and the specific detection of putrescine was realized.(1)This thesis has synthesized three new types of alkynes with high activity terminal alkynes through formylation reaction,Grignard reaction and oxidation reaction based on1,4-Dimethoxybenzene,pyrene and carbazole.Their structures were characterized by 1H NMR.(2)The effects of different solvents on the compounds were studied by UV-Vis and fluorescence spectra.The results show that the three compounds have good fluorescence emission performance in DMSO.(3)The selectivity of the compounds to biogenic amines was analyzed by the fluorescence method.The three compounds have good selectivity to putrescine.The fluorescence intensity of the compounds decreased with the increase of putrescine concentration.The detection limits were 3.18×10-7 mol/L,5.96×10-7 mol/L and 4.5×10-7 mol/L,which were better than the national food safety standard.(4)The possible detection mechanism was speculated by 1H NMR,which was consistent with our expected click reaction mechanism.(5)The standard addition recovery experiment shows that the three compounds have been successfully applied to the detection of putrescine in beer samples,and had good application space and development prospects.
