To determine the contents of 10 organic acids of oxalic acid,quinic acid,tartaric acid,malic acid,ascorbic acid,lactic acid,acetic acid,citric acid,succinic acid,and fumaric acid with RP-HPLC from Xinjiang wild apricot.Single factor experiment and orthogonal experiment design were used to optimize and screen the optimum conditions for the separation and purification of citric acid from wild cherry apricot pulp with 717 anion exchange resin.Using Vc as a positive control,To investigate the ability of scavenging the hydroxyl radicals(·OH),superoxide anion radicals(O2-·),and1,1-diphenyl-2-trinitrophenylhydrazine free radicals(DPPH·)and the effect of total organic acids(TOAWA)of wild apricot on lipid peroxidation in vitro;To sthdy the effect of total organic acids(TOAWA)from wild apricot on the changes of lipids and antioxidation indexes in hyperlipidemic rats over time.The organic acid in Xinjiang wild apricot was extracted by ultrasonic using 60%methyl alcohol.The separation was performed on a Inertsil ODS-SP C18 column(4.6 mm×250 mm)at 30℃,mobile phase consisted of methyl alcohol-(NH4)H2PO4(p H2.4)(3:97,v:v)at folw rate of 0.5 m L/min.The response signal was detected at 210 nm.Single factor experiment and response surface design were used to optimize and screen 717 anion exchange resin to separate and purify citric acid from wild apricot.Using Vc as a positive control,To investigate the ability of scavenging the hydroxyl radicals(·OH),superoxide anion radicals(O2ˉ·)and 1,1-diphenyl-2-trinitrophenylhydrazine free radicals(DPPH·).Vitamin C was used as positive control and thiobarbituric acid method was used to observe the lipid peroxidation activity of the lipid peroxidation system of yolk lipoprotein(PUFA)and the inhibitory effect on the lipid peroxidation induced by spontaneous lipid peroxidation,CCl4,Fe2+-Vc and H2O2 in mice liver in different concentrations of TOAWA.180 experimental rats were fed with high-fat emulsion(male and female half),Xuezhikang positive control group,and wild apricot organic acid high,medium and low dose groups.The effects of TOAWA on serum lipid levels and antioxidant index in 20 d,40 d and 60 d experimental rats were detected by kit.10 organic acids chromatagraphic peak was obtained with good separation.The recovery was in the range of93.20%~108.26%and the relative standard deviation(RSD)less 3%,the limit of detection was in the range of 0.01~0.73μg/m L,the limit of quantiation was 0.03~2.39μg/m L.The optimum conditions for the separation and purification of citric acid from wild apricot by anion exchange resin were as follows:The 717-type resin has good adsorption and desorption properties of citric acid components in the wild apricot.The elution flow rate was 0.5 BV/h,the elution concentration was 0.5mol/L,and the p H was 4.Wild Apricot total organic acids had a certain ability to scavenge hydroxyl radicals but was weaker than Vc.When the mass concentration reached 4 mg/m L,the clearance rate of total organic acid scavenging superoxide anion radicals was 56%,which was comparable to that of Vc group;When the concentration of organic acids was 8 mg/ml,the clearance rate reached 97%,and its DPPH·clearance ability was slightly stronger than that of Vc.TOAWA can significantly inhibit the lipid peroxidation activity in PUFA peroxidation system.It can reduce spontaneous liver lipid peroxidation,CCl4,Fe2+-Vc and H2O2 induced hepatic lipid peroxidation in mice,in addition to the inhibition of lipid peroxidation in the PUFA peroxidation system,regardless of dose,other inhibitory lipid peroxidation activities showed a significant dose-dependence.TOAWA significantly(p<0.01)reduced serum lipid levels in hyperlipidemia rats:triglyceride(TG),total cholesterol(TCH),low density lipoprotein cholesterol(LDL-C),and significantly(p<0.01)improved the high-density lipoprotein cholesterol(HDL-C)content,There was no dose-dependent,but a certain time-dependent;TOAWA could increase the activity of lipase(LPS),total lipase(LPL-HL),but there was no dose-time dependence;TOAWA could significantly(p<0.01)enhance the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)and decrease(p<0.05)the serum MDA content with dose and time-dependent.10 organic acids chromatagraphic peaks were obtained with good separation.The method of extraction on organic acid in TOAWA indicates that ultrasonic extraction is more convenient than reflux.The 717-type anion exchange resin was used to separate and purify the citric acid component of wild apricot.Its comprehensive performance was good and it had a good application prospect.Wild Apricot total organic acids have free radical scavenging ability and strong inhibition of lipid peroxidation activity in vitro.TOAWA could reduce serum lipid possibly by reducing serum lipidblood fat levels in hyperlipidemia rats,increase HDL-C concentration and enhance the activity of the body antioxidant enzymes decomposition of triglyceride enzymes.
