In edible vegetable oil as the research object of this research,the optimization of aflatoxin B1 enzyme-linked immune detection methods of pretreatment,further more the pros and cons of different brands of kits,it is concluded that the optimization of pretreatment method with high stability and good accuracy kit,at the same time with ultra high performance liquid chromatography-tandem mass spectrometry comparing,it is concluded that the feasibility of this research,provide the basis for later of edible vegetable oil testing.The enzyme linked immunoassay(elisa)was used to detect aflatoxin B1 of various edible vegetable oils,and the statistical analysis of aflatoxin B1 contamination of edible vegetable oils from different regions and different production dates was carried out with the data of mass detection.The conclusions are as follows:Using ultra high performance liquid chromatography-tandem mass spectrometry instrument to investigate the authority recovery,choose the optimal pretreatment conditions,using the optimal pretreatment conditions using elisa kit for testing of all brands,and the recommended pretreatment conditions with each kit,by selecting the optimal pretreatment conditions incorporated into each kit as general pretreatment method of kits,to optimize the integration of enzyme-linked immunoassay.For the optimization and integration of elisa pretreatment,70% methanol aqueous solution was used,no auxiliary extraction solvent was added,the ultrasonic power was 60 W,and the extraction time was 20 min.The remaining pretreatment methods were optimized and centrifuge at 6000r/min for 10 min,filtrate the absorbent through 0.22 m needle filter head as the optimal elisa pretreatment.The stability,accuracy and practicability of enzyme linked immunoassay(elisa)for the detection of aflatoxin B1 were improved by using ultra-high performance liquid chromatography-tandem mass spectrometry as the reference ratio.The elisa kits with more accurate and stable results were selected by comparing the differences among the kits.Under the optimized experimental conditions,the elisa showed a good linear relationship in the concentration range of 0 ? 50 g/L,with a linear coefficient of 0.9993 and a detection limit of 1.0 g/kg.The optimized elisa was suitable for the quantitative analysis of aflatoxin B1 in vegetable oil.Aflatoxin B1 of various edible vegetable oils was detected with the integrated and optimized method,and the aflatoxin B1 contamination of large quantities of edible vegetable oils in different regions and different production dates was statistically analyzed with the data of mass detection.The conclusion is as follows: in big cities,most of the edible oils in supermarkets and grain stores are stereotyped and packaged by well-known brands,so the quality can be guaranteed.In addition,it can be inferred from the results of canteens and restaurants that in school canteens and large restaurants,more safe edible vegetable oil will be chosen.In a few medium and small restaurants,the edible vegetable oil of great majority restaurant still is relatively safe,just a few medium and small restaurants are for profit,buy bulk peanut oil or the vegetable oil with substandard quality,cause aflatoxin B1 to exceed bid.For such small and medium-sized restaurants,the government needs to strengthen supervision to ensure the safety of edible vegetable oil.Selection of edible vegetable oil,should avoid to choose bulk and squeeze peanut oil,such as peanut oil due to environmental hygiene not effectively monitoring,without finishing process removal of aflatoxin B1,more prone to excess aflatoxin B1,buy in the supermarket stereotyped packaging and choose the famous brand of edible vegetable oil,edible vegetable oil better ensure the safety performance.
