| This paper was aimed at investigating the accumulation of total phenolics(TP),total flavonoids(TF),rutin and quercetin in buckwheat sprouts treated with slightly acidic electrolyzed water(SAEW)and tap water was used as control.Antioxidant indices were evaluated.Besides,the effect of SAEW on the morphological parameters of buckwheat sprouts was also assessed prior to phenolics analysis.The proteomic analysis of buckwheat sprouts was conducted to gain a deeper understanding of the mechanism of their growth and flavonoids accumulation.Meanwhile,systematic application of SAEW was performed and its microbial reduction efficacy was investigated.The fluctuation of p H of SAEW had little impact on the germination rate,sprouts length and hundred – grain weight.SAEW with lower ACC(10 mg / L)could trigger higher germination rate,sprouts length and hundred – grain weight.By contrast,that with higher ACC(30 mg / L)showed significantly adverse effect on the increase of hundred – grain weight.SAEW treatment effectively reduced the total bacteria count and yaests and molds count attached to buckwheat seeds and sprouts when it was applied to seed soaking,germination and sprouts washing.Changes in ACC had no marked influence on the disinfection efficacy during germination.SAEW exhibited equivalent and even better diainfection efficacy than acidic electrolyzed water or Na Cl O solution in each process.SAEW treatment made the TP,TF,rutin and quercetin content rise to 32.27,23.97,19.96 and 1.11 mg / g,respectively.Among SAEWs,the one with lowest available chlorine concentration(ACC)treatment resulted in the highest level of the four indexex..SAEW treatment made the antioxidant capacity of buckwheat sprouts significantly stronger than control after 7 days of germination.Furthermore,buckwheat sprouts treated with SAEW(ACC 10.94 mg / L)had the highest scavenging percentage of DPPH and ABTS,reducing power along with total antioxidant ability(89.64 %,89.80 %,90.50 % and 89.98 %,respectively).SAEW treatment(ACC 10.94 mg / L)made the α – glucosidase inhibitory activity reach 78.88 %.The phenomenon that SAEW promoted their growth is closely associated with the expression of proteins related to energy metabolism and response to oxidative stress.Among them,changes in isocitrate dehydrogenase [NAD] subunit,phosphoenolpyruvate carboxylase and glyceraldehydes – 3 – phosphate dehydrogenase played critical role.Besides,abundance of antioxidant enzymes(especially superoxide dismutase)and other functional proteins(e.g.thioredoxin)was markedly correlated with sprouts growth.SAEW increased flavonoids accumulation in germinated buckwheat by modulating regulation of numerous proteins(e.g.anthocyanin synthase and anthocyanidin reductase)in phenylpropanoid biosynthesis pathway.The application of SAEW with suitable ACC would be bound to promote the flourishing of functional sprouts vegetables industry. |
