Calliptamus italicus and Gomphocerus sibiricus are two dominant species in desert,semi-desert steppe and alpine and semi-alpine steppe habitats in Xinjiang,CLIMEX model was used to predict the distribution and change of the suitable area of the C.italicus and G.sibiricu in Xinjiang under the climate change condition.Secondly,transcriptome sequencing and fluorescence quantitative PCR were used to study the regulatory pathways and functional genes associated with high temperature stress,and the Hsp70 gene was cloned from the C.italicus,the MIOX gene,which is highly expressed in the G.sibiricu,elucidates the difference of the heat adaptation ability of the locust in different areas from the molecular point of view:(1)The CLIMEX model prediction results show that the suitable area of the C.italicus is larger than G.sibiricu under historical climate conditions,and the suitable areas of the two types of locusts are concentrated in the northern Xinjiang region;In the future climate warming scenario,the suitable areas for C.italicus and G.sibiricu will increase.Under the A1 B emission scenario,the suitable areas will move northward significantly.The suitable height area of C.italicus will increase in Ili Kazakh Autonomous Prefecture,Altay and Hami.The area of the G.sibiricu increased in the height suitable area of Ili Kazakh Autonomous Prefecture and Bortala Mongol Autonomous Prefecture.(2)The results of transcriptome prediction and analysis showed that 24 m RNA sequences of male and female atractomorpha and Siberian Atractomorpha(Cinf?Cinm?Gsnf?Gsnm)and male Atractomorpha(Cihf?Cihm?Gshf?Gshm)under suitable temperature and high temperature stress were 1,a total of 258,970,772 raw reads,2 sets of RNA mixtures were sequenced and 25.39 Gb and 22.68 Gb of data were generated from the two sets of RNA mixtures.14,293 and 14,485 transcripts were generated,of which N50 values were 3,161 and 3,600 bases.The male and female locusts identified 37 and 89 DEGs,and the male and female G.sibiricus identified 34 and 52 DEGs.The differentially expressed genes of male and female C.italicus were enriched in 50 and 57 pathways respectively,and the differentially expressed genes of male and female G.sibiricus were enriched in 52 and 35 pathways respectively.(3)qPCR confirmed the expression level of 6 genes in male and female C.italicus and G.sibiricus.Under high temperature,both male and female C.italicus and G.sibiricus upregulated heat-related genes including Hsp70,PDPK1 and CRYAB,in which the expression of Hsp70 was significantly higher than that of other genes,and the expression of Hsp70 in female locusts was higher than that in male locusts.(4)Two highly expressed genes,Hsp70 and MIOX,were cloned and the CDS sequences with lengths of 1076 bp and 879 bp were obtained.
